OBJECTIVE : To observe the effects of hydrodysable tannin: corilagin(Phy-13) and 1,6-di-O-G- β -D-glucose(Phy-16) extracted from Phyllanthus emblica on the adhesion of endothelial cell to monocyte and proliferation of vascular smooth muscular cells(VSMC) induced by oxidized low density lipoprotein(ox-LDL). METHODS: After incubated with Phy-13 and Phy-16, the endothelial cell(ECV-304) culture with OX-LDL. First we test the concentration of MDA of culture medium, then we add monocytes, finally we test the ratio of endothelial cell adhering to monocytes . After cultured with Phy-13 bid Phy-16, VSMC was cultured with OX-LDL. Then we test the proliferation of VSMC with MTT. RESULTS: OX-LDL can increase the ratio that monocyte adhere to ECV-304. Both Phy-13 and Phy-16 significantly decrease the ratio that mononuclear cell adhere to ECV-304, also the ratio decrease with the concentration dependent. The OD that VSMC cultured with ox-LDL group is higher than that of LDL group, and the absorbence is concentration dependent with the compounds. We get the same result when we exclude the OX-LDL. CONCLUSON: The results suggest that Phy-13 and Phy-16 decrease the effect that monocyte adhere to endothelial cell induced by OX-LDL, this result maybe is that Phy-13 and Phy-16 can prevent the endothelial cell from damage by OX-LDL. But both Phy-13 and Phy-16 can promote the proliferation of VSMC, the detail mechanisms is worth to being studying.
