Objective To investigate the genetic instability In transitional carcinoma(TCC) of the bladder by Inter-Simple Seguence Repeat PCR analysis to evaluate the role of genetic alteration to early diagnosis of TCC.Methods Loss of heterozygosity (LOH) and microsatellite instability(MSI) were studied in the tumor tissue and urine sediment of 30 patients,with their peripheral bloods as control, by means of PCR amplification of a panel of seven (CA)n/(TG)n primers as recommend by a recent workshop on genetic alteration in colorectal cancer. 6% denaturing polgacrilamide gel eletrophoresis were performed ,followed by silver staining. Resides The output of PCR ampliation were a set of approximately 20 products ,The DNA sequences were 100-1200 bp . Gains, losses and intensity changes were showed. The instability index was significantly correlated with the tumor grade and stage. In the majority of cases ,the results of tumor tissue analysis were identical with the results of urine sediment analysis.Conclusion 12.8% of genomic alteration in transitional cell carcinoma of bladder and urine samples are different with normal genomic. The more difference genomic alteration was, the higher the tumor grade and stage were .the analysis of genomic instability in urine sediment should be further evaluated as a method for bladder cancer screening in a high -risk group.
