Objective To detect expression of P14ARF protein, P27kip1 protein , p53 protein and explore three proteins' relationship in the genesis and development of human glioma. Furthermore, to investigate the deletion of P14ARF gene in different grade gliomas.Method Expression of P14ARF protein, P27kip1 protein, p53 protein and the deletion of P14ARF gene were detected by using S-P immunohistochemical technique and in situ PCR respectively in 31 cases of brain glioma with different pathological types and 5 cases of normal brain tissue.Result The expression rate of P14ARF protein, P27kip1 protein and P53 protein in low and high pathological grades was 87. 5% and 30. 43% (P<0. 05) , 75% and 21. 74% (P<0.05), 12.5% and 69.57% (P<0.05). A significant negative corelation (r=-0.571 , P<0.01) was observed between the P14ARF protein and pathological grades , the same as P27kip1 protein (r=-0. 521,P<0. 01). On the contrary, mutant P53 protein showed a positive corelation with pathological grades (r=0. 472, P=<0. 01) . The expressionlevels of P14ARF protein and P27kip1 protein were higher with lower grade of malignancy. But the expression lever of P53 protein increased parallel with the elevation of glioma malignant degree. And there was a significant corelationship between the three kinds of proteins (P<0. 05). The deletion rate of P14ARF gene was 12.5% in grade â… , â…¡, 30% in grade â…¢, 53.85% in grade IV. And negative express rate of P14ARF protein was 12. 5%, 50% and 84.62% respectively in â… , â…¡, â…¢, â…£ grade. The changes in DNA deletion and negative protein level were without one to one relation. These deletion rates were related to the increase of malignant grade of brain glioma significantly.Conclusion This study suggests that the expression of P14ARF protein, P27kip1 protein and p53 protein play very important roles in the genesis and development of human brain glioma and they may coexist in it. The abnormal expression of the three proteins cause disorder of cell cycle modulation and increase of cellular proliferation activity which is an important factor to influence the biological character of brain glioma. Inactivity of P14ARF gene initiates the genesis of glioma and leads to malignancy.
