Objective 1, To investigate the expression of CCK receptor and serum CCK level in patient with gallstone. 2 , To investigate the CFTE mRNA expression and the relationship between CFTE mRNA expression and the change of gallbladder motility.Methods We studied the gallbladder emptying function by B ultrasonography on 40 patients with gallstone and 10 patients of control group, and we studied CFTR mRNA and CCK-R mRNA expression by reverse transcript polymerase chain reaction (RT-PCR). Radioimmunoassay was used to explore the CCK level of serum.Results CFTR mRNA expression was higher significantly in test group than in control group(0.513±0.258 vs 0.264±0.105 P<0.01). The gallbladder motility was significantly impaired in test group. The high expression of CCK-R mRNA in test group was lower significantly than that of control subjects(0.59±0.03 vs.0.91±0.04, P<0.01) and the CCK level in serum in test group was higher significantly than that of control subject(P<0.01). The high expression of CFTR mRNA was correlated with CCK-R mRNA expression and also correlated with emptying rito in patients with gallbladder stone, the change of expression of CCK receptors was also negatively correlated with serum CCK level (P<0.01).Conclusion The expression of CCK-R mRNA in patients with gallstones decreased singnificantly and CFTR mRNA expression washigher significantly. The change of expression of CFTR mRNA was correlated with CCK-R mRNA expression and also correlated with emptying rato in patients with gallbladder stone. The higher CFTR mRNA expression play a important role during the formation of cholesterol gallstone by changing gallbladder motility .
