Abstract/Summary:
Aim: To further investigate the anti-tumor effect of the effective composition of Lysimachia clethroides Duby and its mechanism. Methods: Using the active tracking method to find the effective composition of LCD. Then we investigated the inhibitory effects of ZTF on the transplantable tumor models of S180 and H22. Cytotoxicity of ZTF on HL-60 and K562 cell lines was dertermined by MTT and Tryben assay. We used the imrnunohistochemical study, electron microscope observation, flow cytometry, Hoechst kit, agarose gel, RT-PCR and western blot analysis to further investigate the anti-tumor mechanism of ZTF. Results: The ZTF had a significant anti-tumor effect in vivo and vitro. In mechanism research we found that ZTF could significantly reduce the expression of PCNA, and it could induce apoptosis in HL-60 cells. We also made it clear that the ZTF could upregulate the expression p21 and p27 gene, downregulate the expression cyclin B1 gene and c-Myc oncoprotein. ZTF also could block the cell cycle at G2/M phase in HL-60 cells after 48h. Conclusion: ZTF was the effective composition of LCD, and its mechanism was associated with the regulation of several functional gene and protein.
