ObjectivePlatelet plays an important role in thrombosis. Only activating platelet can have biological behaviors such as adhesiveness, deformation, aggregation and release. Compared with silent platelet , activating platelet has changed significantly in membrane glycoproteins, especially changes in PAC-1 and CD62P. PAC-1 is fibrinogen's receptor. It causes platelet aggregation by combining with fibrinogen. So the formation of PAC-1 is the necessary condition for platelet aggregation. CD62P plays an important role in thrombosis and inflammatory reaction by causing adhesiveness and aggregation of neutrophil and monocyt with platelet or endotheliocyte. CD62P is one of the marks of platelet activation because the amount of CD62P on activating platelet membrane could add up to more than 15 times. Some data indicated that platelet activation was closely related with cerebrovascular disease. Carotid atherosclerosis is an important risk factor of cerebrovascular disease. There are many mechanisms how carotid atherosclerosis leads to cerebrovascular disease, but it is still not very clear. We observed the level of platelet activation and analysed the relationship between platelet activation and carotid atherosclerosis in cerebral infarction patients in order to investigate how activating platelet plays role in the onset of cerebral infarction with carotid atherosclerosis, so as to look for new methods for preventing the occurring and reoccurring of cerebral infarction and for intervening inthe deteriorating course of cerebral infarction.Method1. Subject(1) Cerebral infarction group: We examined 76 patients with ahterosclerotic cerebral infarction. They were diagnosed by head CT or MRI. They were patients hospitalized within 72h after they were attacked by cerebral infarction. Patients with vertebrobasilar cerebral infarction were excluded. 39 patients were males, and 37 patients were females. The rang of age was from 45 to 74, the average age was 61.1 7.8.(2) Control group: there were 53 health examination people. They were also examined by head CT or MRI whose results showed that they weren't cerebral infarction patients. 26 cases were males, and 27 cases were females. The rang of age was from 44 to 72, the average age was 59.3 7.6 .Subject with diabetes, hyperlipemia, heart disease, lung disease, liver disease, kidney disease, hematonosis, infection or tissue damage was excluded. Subject who had taken medicine such as aspirin, ticlopidine, agacore or heparin and so on in recent two weeks was excluded.2. Method(1) PAC-1 and CD62P detection: Fluorescence antibodies of CD61 PerCP, CD62P PE, PAC-1 FITC (each 20 l ) and whole blood (5 l ) were put into the experiment test tubes , and fluorescence antibodies of CD61 PerCP, IgGl PE, PAC-1 FITC (each 20 l ), RGDS (10 ul ) and whole blood (5 l )were put into the contrasting test tubes. Let them react under some conditions so that platelets would be dyed by fluorescence antibodies. Then we detected them by flow cytometry.(2) Carotid examined by color Doppler Ultrasound: We examined vertical and transverse section of carotids continuously from bottom to top along the trend of carotid by color Doppler Ultrasound , and carefully checked carotid internal-media thickness, plaque and plaque's locus, form, dimension and character.According to ultrasonic examination, we divided cerebral infarction group intoone with carotid atherosclerosis group and another without carotid atherosclerosis group , and divided control group into one with carotid atherosclerosis group and another without carotid atherosclerosis group.Results1. The relationship between positive rates of PAC-1 and CD62P and cerebral infarction as well as carotid atherosclerosis: Positive rates of PAC-1 and CD62P in the cerebral infarction groups no matter with or without carotid atherosclerosis were significantly higher than those in the corresponding control groups.
