PrefaceScoparone (6,7 - dimethoxycoumarin) is an active principle isolated from traditional Chinese herbs Artemisia scoparia or A. capillaries thumb. As a pure compound, it has cholagogue, hepatic protection, analgesia, hypolipimia, im-musuppression, hypotention and so on.Bronchial asthma is a chronic inflammatory disease of the airways, and characterized by massive infiltration of eosinophils and airway hyperreactivity (AHR) , which are caused by overproduction of inflammatory factors. It was reported that scoparone possessed anti - inflammation and calcium antagonistic effects. We found that the dilatory effect of scoparone on the trachea! smooth muscle of guinea - pig, hence we presumed that scoporone might cure bronchial asthma. There has not been any report about anti - inflammation activity of scoparone on asthma, so we investigated the anti - inflammation activity of scoparone via using asthmatic models of guinea pig.Materials and Methods1. Drug and reagentsSco isolated from the flowers and buds of Artemisia Scoparia Shenyang was fonud that the purity exceeded 99%. All other reagents were analytical grade.2. AnimalsGuinea pigs weighing about 150 -200g were used in this study. (Laboratory Animal Center of Liaoning Medical University)3. Preparation of asthmatic guinea - pig modelsAfter intraperitoneally injecting with 1ml 10% ovalbumin for sensitized, the guinea pigs inhaled 1% aerosol ovalbumin three times as well as control group inhaled equivalence aerosol saline. It showed the success for asthmatic models in appearing dyspnea, rhythmic contraction of abdominal muscles. 340mg/L , 42mg/L and 5mg/L Sco was inhaled 30mins before every inhalation of aerosol ovalbumin as well as the control group inhaled the equivalence saline. Those failures of asthmatic models were eliminated.4. Determination of plasma and HALFUnder the anesthetic condition (Urethane, 1.25g/kg, Ip. ) , the blood collected from the hearts and about 1mm lung tissue was ligated and harvested from the edge of lung after 24 h from the final champion. Perfuse 5ml sterile saline into the treachea via the bronchial catheterization for 3mins and collect the bron-choalveolar lavage fluid (BALF) for tow times.5. Preparation of lung tissueThe masses of fresh lung tissue were fixed for Ih in the mixture of 0.1 mol/ L dimethyl arsinic acid sodium salt buffer solution(pH7.2, DAASSBS) and 2. 5% glutaraldehyde, rinsed three times by using 0. 1 mol/L DAASSBS ( pH7. 2). Thereafter the samples were tailored into frozen sections. The above frozen sections were rinsed five times by using 0.1 mol/L DAASSBS ( pH7.2 ) after incubation 10h at 37 C. The samples were finally fixed by using 1% AsO4 buffer solution (pH7. 2) after dipping in 0.15% silver nitrate solution for 10min. After rinsing lOmins in 0. 1 mol/L DAASSBS (pH7. 2) , the samples were dehydrated by using propioic aldehyde, embed by using Epon, sliced into ultrathin sections and dyed by using acetic acid uranium. The above sample sections were observed via image pattern analysis.Results1. The centents of II - 5 in plasma and D ALF of asthmatic models were higher than blank control group, ( P < 0. 01) ; Sco (42mg/L,340mg/L) group can significantly depressed The centents of IL - 5 in plasma and DALF , (P <0. 01).2. The centents of PLA2 in lung tissue of asthmatic models were higher thanblank control group, ( P < 0. 01) ; Sco (42mg/L, 340mg/L) group can significantly depressed the contents of PLA2 in lung tissue( P <0.01).3. Comparing with asthmatic models , the centents of PLA2 in lung tissue and IL -5 in plasma and DALF dont have significant difference in 5mg/L Sco group.DiscussionBronchial asthma, as an allergic inflammation disease, is characterized by massive infiltration of eosinophils caused by overproduct of various inflammatory factors. The secondary mediators play an important role in allergic asthma, such as leukotrienes, prostaglandin and platelet activating factor.
