ObjectiveTo observe the affect of injecting danshen in different time on the protecting effect of hepatic ischemia - reperfusion injury via rat experiment.MATERIALS AND METHODS1. Experimental materials: (1) Experimental animals : Healthy male Wistar rats weighing between 250 and 300 g, supplied by experiment center of CHINESE MEDICAL UNIVERSITY. (2) Drug and reagent : Danshen was supplied by CHIATAL QINGCHUNBAO PHARMACEUTICAL CO. LTD. MDA and SOD testing box was supplied by NANJING JIANCHENG BIOLOGY PROJECT GRADUATE SCHOOL.2. Methods2.1 Grouping : all rats were randomly divided into 5 groups,8 rats each group. Food was fasted 12h before experiment , drinking freely. 1. 25% thiopen-tal(20 -40mg/kg) was injected into abdomen to anesthesia. The model of hepatic ischemia - reperfusion injury was made by placement of a artery clamp at the crotch level of the hepatic artery and vein to interrupt the blood supply of lo-bus hepatis sinister vein and artery for SOmin, reflow initiated by removing the clamp for 120 min. blood and tissue sample was taken to examine.1) control group: animals subjected to anesthesia and laparotomy only, each index was examined after 150min.2) ischemia - reperfusion group: the artery and vein of lobus hepatis sinister was clamped for 30min,each index was examined 120rnin after removing the clamp.3 ) Danshen group I : danshen was. injected into blood via right kidney vein,3g/kg. the artery and vein of lobus hepatis sinister was clamped for 30min, each index was examined 120min after removing the clamp.4 ) Danshen group II : danshen was injected into blood via right kidney vein (3g/kg) when the artery and vein of lobus hepatis sinister was clamped for 30min,each index was examined 120min after removing the clamp.5) Danshen group IH : the artery and vein of lobus hepatis sinister was clamped for 30min, danshen was injected into blood via right kidney vein (3g/ kg) after removing the clamp for 30min, each index was examined 120min after removing the clamp.2.2 Experimental index examination1) Hepatic function examination: Blood samples were processed from left ventricle to determine plasma aspartate transaminase ( AST) ^ alanine transami-nase(ALT) and lactate dehydrogenase (LDH).2) MDA and SOD examination; Liver tissues were taken after I/R and were dipped into normal saline . Liver tissue even serum was made by high speed de-condentrator and was at odds with the community at low temperature . Upper clear fluid was taken and saved in refrigerator ( - 30 Ti ) to be examined.3) Pathology sample: bit hepatic tissue sample was taken from left lobe after reflowing for 120min and was immersed into the 10% formalin . Dyeing by HE, histology alteration was observed by optics microscope.2. 3 Result analysis;all the data were analysed by independent samples T test through the spss!2. 0 Statistics software package.RESULTSCompare with ischemia -reperfusion group, ALT, AST,LDH. MDA in danshen group I , II decrease and SOD rise up ( P < 0. 01 ) , compared with the control group, the index in I/R group had significant difference ( P < 0. 01). The index in danshen group IH had no significant difference compared with the I/R group (P > 0.05) ,the index between danshen group I and II had no significant difference (P > 0.05) ,and there is correspond change in pathology.CONCLUSIONInjecting danshen before ischemia - reperfusion and between ischemia and reperfusion can protect liver from warra ischemia - reperfusion injury . Injecting danshen 30 mintutes after reperfusion has no this effect.
