The Experiment Of The Advancing Chemosensitization By Herceptin

Herceptin is a kind of anti-tumor drug which is monoclonal antibody and aim at oncogene. Herceptin has good effect to metastasized breast cancer patients whose human epithelia growth factor receptor-2(HER-2) are positive. Herceptin is the first treatment drug which aim oncogene at a target treating metastasized breast cancer patients whose HER-2 are positive. It can be used singly and also can be used combining with other chemical therapy drug. In this study, we investigated the effects of Herceptin on drug sensitivity of DDP, ADM and TXT with MTT assay. The main results are summarized as follows:1.HER-2/neu antibody(Herceptin) had obviously inhibition to breast cancer cells 543.2.DDP, ADM and TXT all had inhibition to human breast cancer cells, especially ADM and TXT . When ADM cultured with human breast cancer cells 543 for 6 hours, 30% inhibition rate was 1.0ug/ml. When DDP cultured with human breast cancer cells 543 for 6 hours, 30% inhibition rate was 5.9ug/ml. When TXT cultured with human breast cancer cells 543 for 6 hours, 30% inhibition rate was 3.0ug/ml.3.Herceptin combined respectively with DDP, ADM and TXT had better inhibition to breast cancer cells compared with Herceptin singly.4.Adding Herceptin after using DDP for 24 hours, had themost inhibition to breast cancer cells. Adding Herceptin after using ADM for 24 hours, had the most inhibition to breast cancer cells. Adding Herceptin after using TXT for 18 hours, had the most inhibition to breast cancer cells.
