Objective To studying the change of gap junctionalintercellular communication (GJIC) in malignancy and the relativity between the change and chemosensitivity by a newlevel --Cell biology reactive signal adjustive level toestablish a experimental base for a new exact, simple, sensitive and shortcut Chemosensitivity test for chemic remediar drug in bone tumor.Methods we collected the axenic tumor example to be cell plant from 8 bone malignancy patients who were proved by pathology without chemic treatment, after introducted the planted cell into the hole board and added five kinds of drugs(ADM, VCR , DDP , MMC , MIX) which were in common use into it in three final chroma, we tested the resume of GJIC by fluorescence redistrubution after photobleaching(FRAP)and confocal laser scanning microscope (CLSM) and analysed the datum by computer, and contrasted the datum with the result of MTT method in the same condition.Finally, analysed the relativity of the results between the two groups by statistics means.Results In the condition of final chroma was Cmax*5, there were some resume of GJIC in all the five kinds of drugs in the experimental group, there were some sensitivity to the tumor in all the five kinds of drugs in the control group, there were obviously correlation (r>0. 76, p<0.05) between the two groups in ADM>DDP and MTX, there were no markedly correlation between the two groups in other two drugs. In the condition of final chroma was Cmax*l , there were obviously correlation between the two groups in DDP. In the condition of final chroma was Cmax*0. 2, there were no evident resume of GJIC in all the five kinds of druds in the experimental group, there were some sensitivity to the tumor in all the five kinds of drugs in the control group, there were no evident correlation of the results in the five drugs between the two groups. Conclusion At present, there were lots of differentChemosensitivity test of bone tumor, but every one had it's own limitation . the GJIC of bone tumor was restrained and descended and the chemic treated drugs could resume it's restrain and descend. We have gained some satisfying resultsin our experiment by measuring the resume of the GJIC and studying the correlation with the MTT. Because of it's simple, sensitive and shortcut, the GJIC could be a well means to direct the clinic chemic treatment. A studying of more kinds of drugs and large samples was necessary to establish a perfect, criterion and high reliability base for the chemosensitivity test of bone tumor.
