Objective To investigate an appropriate method for the preparation of acellular dermal matrix with high osmotic sal-trypsin and its possible pathogenesis. Methods 4 SD rats were anesthetized and disinfected,the hair of trunk were taken off by 8% barium sulfide.Split-thickness skin,0.4mm thick,was obtained from rat's full-thickness skin with a dermatome and dipped into 37C 1M NaCl solutions for 48h to take off the epidermis,then the cells were disgested by 37 C 0.25% trypsin and0.1%EDTA solutions for 50, 60, 70, 80, 90minutes.The resulting acellular dermal matrix (ADM) was freeze-dried in a freeze-dryer and detected with histology and microbiolog.Results The ADM which was disgested in different time is soft, ductility, extensibility, spring and undisrupted. The epidermic cells were taken off completely, The Basement membrane complex and collagen fibers of ADM which was disgested for 90 minutes were destructed, but thats of ADM which was disgested for 60, 70 minutes are exist normally and the components of cells are exist too.. The Basement membrane complex and collagen fibers of ADM which was disgested for 80 minutes are exist and the epidermis is extinct. There are no bacteria in ADMConclusion The ADM which was obtained from the high osmotic salt-trypsin method has normal Basement membrane complex and collagen fibers but no cells.It's soft, ductility, extensibility, spring and undisrupted...
