Objective: To investigate the effects of human S100A14 gene on human esophageal squamous cell carcinoma cells.Methods: The sense and antisense S100A14 expression plasmids were constructed , then the human esophageal squamous cell carcinomal cell line KYSE-2 was transfected with sense or antisense S100A14 respectively. The growth properties were subsequently observed. Growth of cells were observed daily by MTT assay, the status of cell proliferation were observed by Plate colony formation, and the cell cycle of the cells were observed with flow cytometry .Results: MTT assay for cell growth :The absorbance of the positive cell clones shows sense S100A14 cells >KYSE-2and antisense S100A14 everyday. According to the growth curve we observed that the cells transfected with sense S100A14 grew fastest. There were no obvious difference between the cells transfected with antisense S100A14 and KYSE-2cells.Colony formation assay: Compared with the cells carrying sense S100A14, the antisense S100A14 cells and KYSE-2 cells displayed much smaller and fewer colonies. There are no obvious difference between the antisense S100A14 cells and KYSE-2 cells. Flow cytometry for cell cycle: Compared with the KYSE-2 cells and antisense transfectants, the cell cycle of sense transfectants changed. Conclusion: S100A14 had the ability to enhance the growth properties, and might increase the level of malignancy Of ESCC KYSE-2 cells.
