OBJECTIVE: To detect the deletion of pl4ARF gene and it's expression in human gliomas.Try to put forward a new thinks about carcinogenesis of gliomas and clinic gene diagnosis and gene therapy.METHODS: 68 brain gliomas specimen ranging from different grades were detected by In situ-PCR and Immunohisto Chemisty.RESULTS : The deletion rate of pl4ARF gene was 10. 0% in grade I,II, 21.4% in grade III, 40.0% in grade IV; The negative express rate of pl4ARF gene was 10.0% in grade I,II, 25.0% in grade III, 43.3% in grade IV; The changes in DNA and protein level were without one-one relation. These deletion rates were related to the increase of malignant grade of brain glioma significantly (p<0. 05).CONCLUSIONS: The abnormity of pl4ARF gene cause thedisorder of cell cycle modulation and increase of cellular proliferation activity .It is an important factor to influence the biological character of brain glioma.
