Objective To observe the effects of propofol on myocardial mitochondrial impairment induced by H2O2 ,we constructed anoxic model of isolated mitochondrion from myocardial cells of Wistar rats hearts and detected mitochondrial permeability transion (MPT)and mitochondrial transmem-brane potential( △ψm).Methods Healthy Wistar rats (wight 190-210g)were killed by cervical dislocation.The hearts were quickly removed and cut enoughly. Myocardial mitochondrion were isolated by the method of differential centrifugation. Protein concentration of the mito- chondrial suspension was determined by the Lowry method.Purity and lives of mitochondrion were determined by the succinate dehydrogenase(SDH) lives.In the course of experments,the samples of lower protein concentration(<0.5mg/ml)and lower lives(<5.6± 1.4)were abstained.The experments were divided 3 groups: H2O2 group (100umol/L H2O2);Propofol; H2O2(100utnol/L)+propofol group. The second and last groups were divided 5 groups according the different concentration of propofol:A.25 umol/L propofol B.50 umol/L propofol C.100 umol/L propofol D.150 umol/L propofol E.200 umol/L propofol.We comparedMPT and △ψm in different groups.Results The detection of MPT and △ψm: the data of different times ineach group were compared,the results hadn't statistics significance(P>0.05).Therefore,in different groups comparison,we choosed the data oftime 5 (reacting five minutes)to compare.Determination of MPT:1. Propofol group: 100 umol/L P,150 umol/L P and 200 umol/L P were respectively compared with 25 umol/L P.The results showed that they have statistics significance(P<0.05).At the same time, 150 umol/L P and 200 umol/L P were respectively compared with 50 umol/L P. The results showed that they have statistics significance(P<0.05). While 25 umol/L P was compared with 50 umol/L P,the result hasn't statistics significance(P>0.05).2. H2O2(100umol/L)+propofol group: 50 umol/L P+H,100 umol/L P+H,150 umol/L P+H and 200 umol/L P+H were respectively compared with 25 umol/L P+H. The results showed significantly(P<0.05). At the same time, 50 umol/L P+H, 100 umol/L P+H and 150 umol/L P+H were respectively compared with 200 umol/L P+H. The results have statistics significance (P<0.05).3. Comparison in groups: 25 umol/L P,,50 umol/L P and 25 umol/L P+H were respectively compared with H2O2. The results have statisticssignificance(P<0.05). At the same time, 25 umol/L P+H and 50 umol/L P+Hwere respectively compared with 25 umol/L P. The results have statistics significance(P<0.05), while 50 umol/L P+H was compared with 50 umol/L P,the result has statistics significance(P<0.05).These results show: In the process of MPT determination,MPT data obviously decrease after adding H2O2(100umol/L).When adding H2O2 (100umol/L) with 25 umol/L P or 50 umol/L P,the data are increasing. Therefore,the data are lower than them after adding purely 25 umol/L P or 50 umol/L P.The effect of higher concentration propofol (100,150,200 umol/L)is not obvious. Determination of in:1. Propofol group: 25 umol/L P ,100 umol/L P,150 umol/L P and 200 umol/L P were respectively compared with 50 umol/L P. The results have statistics significance(P<0.05).2. H2O2(100umol/L)+propofol group: 100 umol/L P+H, 150 umol/L P+H and 200 umol/L P+H were respectively compared with 25 umol/L P+H and 50 umol/L P+H, the results have statistics significance(P<0.05). While 25 umol/L P+H was compared with 50 umol/L P+H,the result hasn't statistics significance(P>0.05).3. Comparison in groups: 50 umol/L P, 25 umol/L P+H and 50 umol/L+H were respectively compared with H2O2, the results have statisticssignificance(P<0.05). At the same time,they were respectively comparedwith 25 umol/L P, The results have statistics significance(P<0.05). These results show.
