PART I In vitro culture and identification of dendritic cells (DCs)Objective To study the feasibility of the culture of DC from peripheral blood of patient with HCC in vitro.Methods To establish the culture system of human DC in vitro and to analyse the appearance, Phenotype, harvest rate, purity and function of DC cultured.Results The culture system established successfully in vitro. The characteristics of classical DC were observed. The harvest rate was 12.8±0.13%, purity was 87.5±1.2%, Concentration of IL-12 in supernate was 1.0±0.2ng/ml.Conclusions The ideal DCs both in quantity and function can be obtained via the in vitro culture from blood of patient with HCC,and they can be used as a new concept vaccine for HCC therapy.
