Objective (1)To investigate the effect of curcumin and its derivatives on various cell lines and growth of tumor in mice and discuss the mechanism of it. (2)To investigate the effect of poly(ethylene glycol)(PEG) conjugated curcumin on Sarcocarcinoma(S180) in mice and its probable mechanism.Methods (1)MTT reduction assay was taken to observe the effect of curcumin and its derivatives on proliferation of bovine aortic smooth muscle cells(BASMCs), human umbilical vein endothelial cells(ECV-304) and human stomach adenocarcinoma cell line(SGC-7901) maintained in medium containing 10% fetal bovine serum(FBS).(2)The effect of curcumin and its derivatives on the migration of ECV-304 maitained in medium containing 10% FBS was investigated by agorose assay. (3)The mouse tumor model was used to investigate the effect of curcumin and PEG conjugated curcumin on tumor growth.Results (1)The inhibitory effect on the proliferation of ECV-304, SGC-7901 and BASMCs by curcumin and its derivatives are different. (2)Curcumin and its derivatives can significantly inhibit the migration of ECV-304 induced by FBS. (3)Curcumin and its derivatives and PEG conjugated curcumin have remarkable inhibitory effect on the tumor growth.Conclusion (1)Curcumin and its derivatives can significantly inhibit the proliferation and migration of ECV-304 induced by FBS, thus can inhibit angiogenesis. (2)Curcumin and its derivatives and PEG conjugated curcumin have remarkable inhibitory effect on the tumor growth. The inhibitory effect of PEG conjugated curcumin on tumor excels that of Curcumin and its derivatives.The inhibitory effect is paralleled with molecular weight of PEG.
