Objective: To survey expression rule and function of ICAM-1 on endometrium of early preglant mice during the blastocyst implantation.Methods: Endometrium from day 1 to day 5 in early pregnancy mice were used as follows study: first, expression level of ICAM-1mRNA is determined by RT-PCR. Second, expression level of ICAM-1 is determined by western-blotting. Third, the expression level of ICAM-1 is determined by immunohistochemistry. Fourth, blastocyst implantation rate was registered by injecting monoclonal antibodies against ICAM-1 to the early pregnant mice body.Result:First, expression level of ICAM-1mRNA on endometrium of early pregnant mice begins to increase from day 1-3,day 4 reach the highest and day 5 begins to reduce gradually.Second,result from western blotting is the same with result from RT-PCR.Third,expression level of ICAM-1 were determined by immunohistochemistry from endometrium epithelia,matrixcells,blood vessel epithelia,also the quantity and intensity of expression have certain of rules. Endometria epithelia of day 1 to 2 expressed much higher than matrix cells and blood vessel epithelia but matrix cells and blood vessel epithelia expressed much higher than endometria epithelia of day 3 to 5.Fourth, injecting monoclonal antibodies against ICAM-1 to the early pregnant mice body, blastocyst implantation rate increase, and that is not effect for the late pregnant mice.Conclusion: The dynamic expression rule of ICAM-1 gene and protein show that ICAM-1 participate in the blastocyst implantation. By injecting the monoclonal antibodies against ICAM-1 to the early pregnanct mice, the result prove more that ICAM-1 participate in forming the endometrium endurance to blastocyst implantation.
