Objective To discusss the effect of diclofenac sodium on inhibiting of rabbit corneal epithelial cells(RCECs) and explore it's mechanism. Methods The primary culture and subculture of RCECs were exposed to different concentrations of Diclofenac sodium,Acular(Ketorolac tromethamine),Dexamethasone and Lincomycin in experimental group and to culture media in contrast group. The inhibtive characteristics to the cells' proliferation were measured with methyl thiazolyl thiazolium(MTT) assay. The changes of cells cycles and the apoptosis rate were observed with flow cytometer(FCM). The ultrastructure of cell were observed with light microscope and transmission electron microscope. And the changes of the apoptosis indexes are measured by immunocytochemistry. Results Diclofenac sodium,Acular,Dexamethasone and Lincomycin can inhibit proliferation of RCECs ,while Diclofenac sodium showed strongest antiproliferative effect. After the treatment of 27.27ug/ml—54.55ug/ml diclofenac sodium,RCECs showd the marked inhibition whenever in 24h,48h or 72h(P<0.01)and showed dosage dependence. FCM result showed that the G0/G1 phase cells obviously increased and the apotosis cusp and rate increased. Immunocytochemistry showd wide type p53 and Caspase-3 increased while PCNA weaken in the drug added group. Conclusion 1,Both Diclofenac sodium and Acular has inhibitive effect on RCECs and Diclofenac sodium showed stronger effect. Diclofenac sodium showed more obvious inhibitive effect on RCECs than Dexamethasone did. 2,Diclofenac sodium has obvious inhibitive effect on RCECs and showed dosage—effect relation. Diclofenac sodium may exert it's antiproliferative effect by inducing apotosis and ceasing cells cycles. 3,It should be further studied on the drug's safe dosage scope,ill effect and so on.
