ObjectivesThe primary objective was to investigate whether apigenin can inhibit the produce of NO iNOS and the apoptesis of cartilage in vivo . The secondary ob-jective was to probe whether apigenin can become a promising drug candidate.Methods24 rabbits were averagely divided into 3 groups at random. (1) Persudo -operation (2 ) Operation ( 3) Operation and adding drug. (1) Persudo - operation group: We cutted the skin of right knees asepsisly , stitched immediately. Two weeks later, we injected DMSO(0. 1ml) into the same knees once every week and abdominal cavity twice every week for4 weeks. (2) Operation group: We cutted the anitiror cruciate ligament and patella ligment of the right knees. Two weeks later, we injected DMSO(0. 1ml) into the same knees once every week and abdominal cavity twice every week for 4 weeks. (3) Operation and adding drug group: We cutted the aniterior cruciate ligament and patella ligment of the right knees. Two weeks later, we injected apigenin(15mg/kg) and DM-SO(0. 1ml) into the same knees once every week and abdominal cavity twice every week for 4 weeks. Before operation and killing, we extracted vein blood 2 ml and stored them at 70 C. After operation for 6 weeks, we killed all the rabbits and cutted the right knees . Firstly, we observed the change of cartilage and synovie with naked eyes. We divided the cartilage into three groups, one group was stained and observed by microscope. The other one was observed by trans-elecmicroscope. The last one was detected by flow cytometry.ResultsThe differences of NO values between persudo - operation group and operation group were significant( P<0.01) , the same thing was the differences be-tween persudo - operation group and operation and drug group; Same as operation group and operation and drug group. The differences of iNOS values were the same condition. The differences of the ratio of cartilage apoptesis between operation group and operation and drug group were remarkable.ConclusionsThe experiment proved that apigenin can inhibit the produce of NO iNOS and the apoptesis of cartilage in vivo. Apigenin have the opportunity to become a component of drug to remedy OA.
