Collagen is one of the important and polyfunctional protein from pig skin. This thesis was to choose the optimum extracting conditions of collagen and disassemble it into peptides. And make them combine to the CrCl3.6H2O, Then observed its effects on blcod-glucose and acute heat exposure in the KM rats.Collagen was extracted from pig skin by using hydrochloric acid method and the best extraction conditions were confirmed. The results showed as follows: During the collagen was extracted by 2M HC1. The PH value was sustaining regulated by HC1. The result indicates, the optimum temperature and time were 85 C, 3h. Then make them combine to the CrC13.6H2O.KM rats used as test adopts, by injecting alloxan in the dose of 200mg/kg into the abdomen, diabetes animal models were established. In comparison, normal rats were injected in the same dose of 0.9% sodium chloride solution. After this, those were treated as diabetes model rats whose fasting blood sugar content was above 11.1mol/L. Experimental rats were divided into 4 groups: normal rats, normal rats fed chromium, diabetes rats, diabetes rats fed chromium. Diabetes were raised in the dose of 1ug/d Cr3+ by drinking, 6 subject rats in each group were killed every week, their blood parameters were determined by biochemical reagent kit. After 6 weeks administration, no significant difference can be observed between normal rats blood parameters and those of rats fed chromium(P>0.05); there was an obvious decrease between the blood sugar content and triglyceride of diabetes rats and those of diabetes raised chromium(P<0.05); The livers were taken from each groups randomly to homogenize them, the soluble protein was extracted in the 0.9% sodium chloride solution and deposited in alcohol. After centrifugation, the proteins were dissolved in the sample buffer solution and separated in SDS-PAGE, there was no difference between the protein strips.Fetching 70 rats were divided into 2 groups, among them one group feeds the organic chromium, the method is the same.6 rats of each group were killed every other day, and blood index determines in the same way. After raising for 12 days, there was no difference between normal rats and those feeds organic chromium, though emerging significant difference at the beginning, their livers albumen were electrophoresis in the same method as above, the difference is obvious.Heat stress experiment: Fetching 24 rats separately, the male and female are half and half, divided into 4 groups. The male and female each have one group that feed with the chromium in the dose of 1ug/d, after 4 weeks administration, all rats were placed in the oven in 43 degrees Centigrade all around, In the oven of 30% of relative humidity, the heat tolerance time and the death rate after 1 hour of each rat was write down. The heat tolerance time of female rat was longer than that of male rat, but there was no significant difference between those. The heat- resisting time of female rat feeds chromium was more longer than the other female group(P<0.05,and there was not have remarkable difference between the male groups(P<0.05),Fetch the liver, kidney, heart, leg, and draw the albumen electrophoresis, there is no difference in every group albumen.
