Object i ve Through studying the expressions of P53. CD56 and PTEN in Manlignant Peripheral Nerve Sheath Tumors(MPNST) and analying the DNA ploidy by the flow cytometry from the level of single cell, we explored the relations between them and the biologic behaviors. of MPNST.Method Using the method of immunohistochemistry, we selected 42 cases of MPNST and 23 cases of benign Peripheral Nerve Tumors to examine P53. CD56. PTEN these three indicators, all the samples are paraffin embedded. At the same time, we analyzed the DNA ploidy of all these samples by flow cytometry. The results were tested by statistical method(exact method, rxc chi-square test, spearman' rank correlation).Result P53 , CD56 and PTEN were all expressed differently between benign and malignant Peripheral Nerve Sheath Tumors and the differences were all sign ificant (X2=34. 7191, P>0.001 ; X2=6. 9556, P<0. 01 ; X2=12. 4780, P<0. 001). The stronger expression of P53 was positively related to the degree of the malignancy of MPNST(rs=0. 326, P<0. 05). From the analysis of DNA ploidy, we foundthat the benign tumors were all diploidy(DI # 0.90~l. 10), and there were 4 aneuploidy in 42 cases of malignant tumors, the rate of aneuploidy was 9.5%(4/42), and all the aneuploidy was low aneuploidy(DI 值 >0. 90). There were 6 cases of tetraploidy tumors in the other samples. SPF and PI index were also different between the benign and malignant Peripheral Nerve Sheath Tumors, the differences were significant(X2#28. 674,P>0.001 ; X2 # 23. 730, P<0. 001) ; SPF and PI index are also associated with the histological grade of MPNST(X'=8. 463, P<0. 05 ; X2=7. 824, P<0. 05) and they had positive correlation with the degree of the tumor' malignancy (rs#0. 418, P>0.01 ; rs#0. 347, P>0. 05). PI index was related to the expression of P53, the differences were significant(P#0.0163). The degree of the expression of P53 was positively correlated to the appearence of the aneuploidy. The degree of the expression of P53 was positively correlated to the SPF and PI index(rs#0. 318, P>0. 05 ; rs#0. 354, P>0. 05). Among all of the factors which influent the prognosis, the appearence of the aneuploidy and SPF index associated with the prognosis(P=0. 02174).Conclusion P53, CD56 and PTEN can be used as the worthy factor to estimate the property of benign and malignant Peripheral Nerve Sheath Tumors; P53 can be used as a valuable indicator of to estimate the degree of malignancy of MPNST. From the DNA ploidyAnalysis, SPF and PI index can act as the associated factors to estmate the property of benign and malignant Peripheral Nerve Sheath Tumors. The appearence of the aneuploidy and high SPF, PI index can predict the malignant biological behavior of MPNST.The aneuploidy and SPF index are also the important factors which can influent the prognosis. It is significence that P53 and all the factors of DNA Ploidy Analysis together act as estimating of property of benign and malignant Peripheral Nerve Sheath Tumors and evaluation of the degree malignancy of MPNST.
