Research On Relation Between Inducible Nitric Oxide Synthase (INOS) And Tumor Cells Proliferation,Metastasis And Its Clinical Significance

Objective The expression of iNOS bFGF CyclinD1 erbB-2 were studied by immunohistochemistry(SABC) to investigate the relationships between the expression of iNOS and proliferative index of breast carcinoma such as bFGF CyclinD1 and erbB-2.The relationship between expression of iNOS and ages menstruous status tumor size histological stage TNM stage venous invasion ER PR lymph-node metastasis distant metastasis and survival rate in 5 years.Methods 69 cases of breast carcinoma were studied.iNOS bFGF CyclinD1 erbB-2 were all detected immunohistochemically in formalin-fixed, puraffin-embeded sections. The results of erbB-2 expression were offered by pathology department.Results iNOS protein was expressed in 43 cases.The positive rate of iNOS was 62.3%(43/69),and it did not significantly correlate with age menstruous status histological stage venous invasion ER and PR.The expression of iNOS was statistically correlated with some oncogene proteins and their related protein such as bFGF CyclinD1 and erbB-2(P=0.015,P=0.004,P=0.003).So iNOS can promote proliferation and development of breast carcinoma cells by the way of these genes.The positive rate of iNOS protein(58.7%) in cases whose tumor size <5cm is lower than in cases whose tumor size>cm(69.6%),but there was no statistical significance.However,significant relation was found between the expression of iNOS and TNM stage.The expression of iNOS was also significantly related to distant and lymph-node metastasis(P=0.024,P=0.008),and significant correlation was also found between expression and disease-free period.Survival rates in positive cases was always lower than in negtive ones.Conclusion On one hand,iNOS and its product-NO may stimulate the expression of some ongene and related genes such as bFGF CyclinD1 and erbB-2.Therefore,iNOS and NO can accelarate proliferation of breast carcinoma cells.On the other hand,iNOS and NO can promote lymph-node and distant metastasis.In general,iNOS plays an important role in proliferation metastasis and prognosis of breast carcinoma.
