Objective: The aim of this study is to evaluate the effects of different factors, including different rolling speed, using different shape of burs , number of stops set on handpieces and concentration of the bacterial suspension, on bacterial contamination of handpieces. Methods: Bacterial contamination of 20 handpieces were analyzed in different rolling speed, using different shape of burs, number of stops set on handpieces and concentration of the bacterial suspension. Results: No statistical different (P>0.05) of handpieces inside contamination was observed relevant to different rolling speed, statistical different (P < 0.05) was observed relevant to number of stops set on handpieces, using different shape of burs, concentration of the bacterial suspension. Conclusion: The concentration of bacterial suspension is the key factor effecting the inside contamination of handpieces. The amount of bacterial culture of group handpieces that using round bur is more than that using fissure bur. Number of stops set on handpieces is not the factor that can effect the inside contamination of handpieces.
