[Background] Wound Repairing is the inherit function of human heredity trait during the growth. Wound healing is a complex physiology process, which is known little about how to adjust the process in molecule level. At present, regulation of multi-growth factors and their receptors play important role in the healing process. The deficiency of the growth factor is attributed to the un-recovered wound, and now, the reason that the wound can not be cured is associated with not only the absent of growth factor but also the abnormal activation of matrix proteinases. The proteinases can dissolve extracellular matrix, recede the activatity of growth factor and affect the interaction of growth factor to cell and cell to cell. Therefore, the adjustment between the growth factor and matrix metalloproteinase is the key to protect the stabilization of micro-environment and to accelerate the wound healing. More and more attention were paid to the authentic mechanism of the wound healing. At present, the treatment of the chronic infection ulcers is the heated topic in medicine. Especially under the condition of the humid and hot, the wound is highly susceptible to the infection and easily developed the chronic wound. Many medical workers were puzzled with therapies of the chronic ulcers yet. Therefore, the research of the treatment measure of the chronic wound should become an important topic. Recently, The GF (growth factor) research brings hope to resolve the difficult problem, but there are a lot of difficulties to be overcomed. For example, the MMP-9 quantity in the chronic wound is higher than that in the acute one and the GF quantity is lower than that in the acute wound. Some experts assumed that ectogenesis GF given tothe chronic wound could made the rehabilitate cell resurvival and accelerate the wound rebuilt and down-regulate the level of MMP-9. It was known that MMP-9 degraded ECM and be tuned by GF. But why is that? What is the relationship between GF and MMP-9? Why could the wound plerosis cell get re-survival after ectogenesis GF was applied to the wound? This study will make closely analysis about the relationship between GF and MMP-9 during the process of the wound healing. As a result, some effective therapy and defensive measures will be provided for incurable wounds in clinic.[Objectives] To investigate the expression of MMP-9 and TGF-B in the wound healing and to probe into their function and the relationship during the process of the wound healing. Then to provide theory guide of the clinic precaution and treatment about the chronic war wound.[Methods]The adult rat 36 were divided into the normal group and the humid and hot groups so as to establish the animal models of blasting injury in rats under humid and hot environment.On the basis of the animal model, the adult rat 48 were divided into the normal group and the therapy group after injury under the humid and hot environment and the wound fluid and the wound tissue were collected. The MMP-9 quantity and the TGF-P quantity in the rat wound fluid were investigated by means of zymography and ELISA respectively. The expression of MMP-9mRNA and TGF-PmRNA of the wound tissue were inspected with in situ hybridization. [Results](1)The healing rate was zero in the humid and hot group; nine rats wound were cured in the normal group in the 21d after the formation of the war wound. Thirteen rats were cured in the normal group by the 28d, but only six rats were cured in the humid and hot group. The healing rate in the humid and hot group were compared with that in the normal group, The differencewas significance (p=0.044).(2)The MMP-9 and TGF-p were detected by Zymography and ELISA in the wound fluid. The results showed, that the normal group: the MMP-9 level elevated after the injury and it's climax value was in 48th hour; the climax of the MMP-9 lasted to 5th day, and slowly descented, and in the 14th day, elevated again; the TGF-|3 level attained the apex in 24th -48th hour, and declined smoothly, and reached the climax in 7 days again, and decl...
