Objective: The aim of this study was to investigate the protective effects of 17-beta-estradiol on liver ischemia reperfusion( I/R )injury in male rat model and to clarify its possible mechanism. Methods: ( 1 ) The model of partial liver ischemia reperfusion in male SD rat were performed. Rats were randomly divided into two groups. Control group: N.S was given 1 hour before liver ischemia reperfusion injury at 0.5ml everyone intraperitoneally. Experimental group: 17-beta-estradiol was given 1 hour before liver ischemia reperfusion injury at 4mg/kg intraperitoneally. After 90 minutes hepatic ischemia serum ALT , AST, LDH and tumour necrosis factor-( TNF- ) were analysed statically between two groups at 1 hour 2 hour 4hour, 1 day, 3day, 5 day and 7 day after liver reperfusion and before hepatic ischemia. ( 2 ) Rats which endured 4 hours liver reperfusion were randomly divided into three groups: shame operation group which did not endure liver ischemia, control group which were given N.S 0.5ml intraperitoneally before 90 minutes hepatic ischemia , experimental group which were given 17-beta-estradiol 0.5ml intraperitoneally before 90 minutes hepatic ischemia. The numbers of polymorphnuclear cells ( PMNs ) in liver and the expression of nuclear factor-kB (NF-kB ) in liver were analysed statically among three groups.Results: ( 1 )The levels of serum ALT, AST, LDH and TNF-a in control group were significantly higher than those in experimental group at 1 hour 2 hour 4 hour 1 day 3 day and 5 day after liver reperfusion. The levels of serum ALT AST LDH andTNF- in control group did not fall to normal until 7 day after liver reperfusion, however, those in experimental group were normal in 5 day after liver reperfusion. ( 2 )Liver injury in control group were more serious than that of groups. There were serious liver cells necrosis and central vein congestion in control group. Liver injury in experimental group was a little more serious than shame operation group only. There was no liver injury in shame operation group. The numbers of polymorphnuclear cells ( PMNs ) in liver in control group were significantly higher than that of experimental group and shame operation group. The PMNs in shame operation group were lower than that of experimental group. ( 3 ) There were a little expression of NF-KB in liver in shame operation group. Expression of NF-KB in liver could be seen much both in control and experimental group but that in control group were more remarkable than that of experimental group.Conclusion: ( 1 ) 17-beta-estradiol may be restrain the expression of NF-KB in liver after liver ischemia reperfusion injury. ( 2 ) 17-beta-estradiol significantly protects liver from ischemia reperfusion injury and accelerate comeback of liver function after liver ischemia reperfusion injury.
