Experimental Research on the Function of Schwann Cells to Neural Stem Cells Cocultured in vitro and Their Relative Biocompatibility Objective: To evaluate the function of Schwann cells obtained from mouse peripheral neural myelin on the rat neural stem cells when cultured together in vitro, to certify whether Schwann cells can promote the survive, proliferation, differentiation of the neural stem cells and whether both cells had easier biocompatibility; At the same time, we examined the myelination of the neurons accomplished by the Schwann cells and estimated the neuron-neuron synapses primarily.Methods: The sciatic nerve and the brachial nerve were harvested from 3 3-day-old Wistar rat, and we selected the tissue clot method to culture the Schwann cells successfully. After cultured for one week, the cells were purified by the gradual deposition method, and then we used S100 immunohistochemical staining and microphotography techniques to certify the Schwann cells. The neural stem cells were obtained from the newborn Wistar rat brain and cultured by the normal shifting cells culture method to amplify the quantity. As the certification of Schwann cells, the neural stem cells were tested by the ABC immunochemical method. Then the cells were cultured together by five groups: research group I : NSC+ shifting SC + DMEM/F12; research group II: NSC + adhered SC + DMEM/F12; control group I : DMEM/F12+the medium obtained from SC culture + NSC; control group II: EGF/bFGF+DMEM/F12+NSC; control group III: NSC+DMEM/F12+EGF. And we used the immunohistochemical method to distinguish the differentiation result of the neural stem cells, and we observed the morphological changes of the neural stem cells. Five groups were cultured by the normal method, changing culture medium every 2days and amplifying due to the condition the cell regeneration.Results: During the 2 research groups, the majority of neural stem cellscocultured with Schwann cells not only had larger diameter than the control groups but also protruded several elongate processes. And we found the expression of NF was high, which represented more newborn neurons and fewer other neural cells including the oligodendrocytes and astrocytes etc. While according to the control groups, except the groups III, the other two groups were found little worse to survive than the research group, but not only the number of the NSC but also the proportion and the normal axon length of the new neurons differentiated from the neural sphere had significantly difference, especially the group III where there was almost no differentiation of newborn neurons not along the length of the axons.Conclusion: During the coculture of NSC and Schwann cells, both cells can survive prosperously and we think they have an easier biocompatibility. Besides this the Schwann cells can promote the survival, regeneration and differentiation of the neural stem cells in vitro, and the two different germ cells can establish some contact conjoint like the neuron-neuron synapses.
