Thesis OneThe Study of ATRA Combined With FUDR in the Treatment of Hepatocellular Carcinoma Cell Line SMMC7721Objective: In order to investigate the influence of all trans-retinoic acid (ATRA)and floxuridine (FUDR) on hepatic carcinoma cell line SMMC7721.Methods: ATRA and FUDR had a single or co-effection on SMMC7721 cell line.The apoptosis of SMMC7721 was detected by fluorescence microscope ,electrophoresis and flow cytometry (FCM).Results: ATRA and FUDR could induce the apoptosis of hepatic carcinoma cellline SMMC7721 respectively. The apoptotic number of co-effection was much morethan those of single effection.Conclusion: The influence of ATRA and FUDR on SMMC7721 was inducingapoptosis. The co-effection was more intense than single effection. ATRA was incoordination with FUDR.A new method in the therapy of liver carcinoma wassupplied by this study.key words: retinoic acid; floxuridine; liver neoplasm; apoptosis.Thesis twoThe Study of ATRA Combined With FUDR in the Treatment of HepaticCarcinoma in Nude MiceObjective: To study the influence of all trans-retinoic acid (ATRA) and floxuridine (FUDR) on hepatic carcinoma cell in nude mice.Methods: After the nude mice was inoculated with human hepatic carcinoma cell BL-7402, ATRA and FUDR was injected into sub-axilla of the nude mice singly or combinedly. The carcinoma mass was investigated through microscope, electricmicroscope and immunohistochemical detection.Results: With the treatment of ATRA and FUDR ,the apoptotic body of hepaticcarcinoma cell line BL-7402 was found in nude mice. Immunohistochemicaldetection, showed PCNA expression was decreased, but BAX, C-myc expressionwere increased.Conclusion: Not only ATRA and FUDR could induce the apoptosis of hepaticcarcinoma cell line BL-7402 in nude mice,but also inhibited hepatic carcinoma cellprolifration. The co-effection of ATRA and FUDR may be extended in clinicalpractice.
