Skin is the biggest organ in human body, is a machine to shield with outside materials, has the complicated physiology construction and various important physiology function. When the skin suffers to the serious burns, wound, infection and diabetes unprogressive skin ulcerates etc, it become hard problem to repair and rebuild the big area skin defect. It will be research focus on how to acquire sufficient seed cell in a short time , how to obtain the safe and dependable factor to promote cell in vitro and how to valid infection in skin transplantation.Objective: To establish and perfect the technique of separation and turn purely the skin cell. To creat with characteristic Chinese medicine, obtain skin inducture with effective immunity activity to expand own keratinocyte . By the observes in vitro, to provide the experimental evidence for the building of tissue engineering skin which is safety, anti- infection and easy to use.Method: 1. To obtain epidermal keratinocyte and dermal fibroblast from rabbit skin by 0.38% Dispase II /O. 053% Trypsin-0. 04% EDTA , detecte the keratinocyte with the expression of ?integrin and keratins 19. 2. To Withdraw ShengJiYe and analyse the concentration of ion . organic matter. little chemical element. To select concentration of ShengJiYe by MTT method with keratinocyte and fibroblast. 3. To observe the resolve of gelatin- chitosan. To creat cell- ShengJiYe carrier compound and investigate its influence and003112211)creature compatibility to cell by half-quantitative. MTT. histology and scan-electronic microscope method. 4. To repair the rabbit full-thickness skin defect by cell- ShengJiYe- carrier compound, make histologies observes, investigating the result of repair.Result: 1. The keratinocyte and fibroblast obtained by Dispase II /Trypsin-EDTA are highly pure and have highly vitality, ? integrin and keratins 19 of keratinocyte have positive expression. 2. MTT result of low and inside ShengJiYe density higher than the normal control group. MTT result of high ShengJiYe density lower than the normal control group in ex- period. 3. Result of histology and scan-electronic microscope express that the cell is closely stick to the material and appearance is normal. 4. Skin defect contracts of cell- ShengJiYe-carrier group is quickly than normal control group, new epidermis have normal construction characteristic.Conclusion: 1. The keratinocyte obtained by Dispase II /Trypsin-EDTA is epidermal stem cell, highly pure and have highly vitality. 2. ShengJiYe have effective immunity activity, can enhance the suitability of keratinocyte and fibroblast. 3. ShengJiYe-gelatin-chitosan carrier satisfy the require toward engineering cell material, make cell grow in its surface and bore, validly repress inflammation, promote keratinocyte function. 4. Own cell- ShengJiYe- carrier compound acquire sufficient seed cell in a short time , tissue engineering technique with Chinese medicine can be used to repair full-thickness skin defects in clinic.
