| Many genes participate the development of HCC, which is a complex process associated with many stages. If we hope to realize the mechanism of the occurrence, development, infiltration and metastasis of HCC, it is necessary to analyze the differential expression of HCC-related-genes during its development.Objective: HCC-related-genes of the differential expression were analyzed by the biological informatics technique. The differential expression HCC-related-genes, which mainly included AFP, COL1A2, UBD, FN, Genimin, GPC3, SPINT2, Stathmin, VEGFB and IGFBP3 ,were studied by RT-PCR technique.Further more the relations between their differential expression and clinical pathological characteristics were analyzed. We also hope to carry through the primary analysis for the molecular margin of HCC. At last it is important to establish the multiplex PCR technique in order to detect the multiple genes' expression.Methods: Surgical specimens of HCC have been collected and reserved in ?0℃.One hundred twenty and five positive clones of the differential expression were choosed to sequence and analyzed by the biological informatics technique. The mRNA were extracted from liver cancer and its paracancerous tissue and semiquantitatively analyzed by RT-PCR technique.Results: Firstly, Eighty three genes from one hundred twenty and five clones sequenced and analyzed by the biological informatics technique were confirmed. There are five genes of which associated with cell division, five of which participate the cell signal transduction or communication, eleven of which are associated with cell or organ defence, twenty two genes participate the gene transcription or theexpression of the protein in the cell, eighteen of which are associated with the metabolism, and another fifteen of which are not be classified. Secondly, between liver cancer and its paracancerous tissue of fifty patients the differential expressions of nine high expression genes took on the statistic significance. Thirdly, the differential expression of SPINT2 between liver cancer and its paracancerous tissue were associated with tumor size; the differential expression of COL1A2 were associated with the quantity of the AFP in the peripheral blood and the histological grade of the tumor; the differential expression of FN and AFP gene were associated with the abnormal AFP in the peripheral blood; the differential expression of ten genes were not correlated with the HBsAg . Fourthly, we detected the different distance tissue around the surgical margin of twenty five patients, ten genes manifested obvious differential expression these tissue. Fifthly, we established the method to detect the genes' expression by the multiplex PCR.Conclusions: Firstly, after analyzing sequences of one hundred twenty and five clones, Eighty three genes of the high expression correlated with HCC were confirmed. Secondly, nine genes of the high expression manifested obvious differential expression between the cancer and its paracancerous tissue. Thirdly, ten genes manifested obvious differential expression among the different distance tissues around the surgical margin. Fourthly, the four genes including AFP, COL1A2, SPINT2 and FN were associated with the tumor size , the histological grade and/or the quantity of the AFP in the peripheral blood .Fifthly, the differential expression of ten genes demonstrated the molecular margin exists around the liver cancer tissue. Sixthly, it was triumphant to establish the method to detect the genes' expression by the multiplex PCR. |
