Objective To investigate the inhibiting effect of Dimethyl Sulfoxide on the proliferation of cultured human retinal pigment epithelial (hRPE) cells.Methods The test was divided into experimental group and control group. The cultured cells of experimental group were treated with DMSO at varying concentrations of 0.5%, 1%, 2%, 4% for 24h, 48h, 72h, 96h respectively.The control added DMEM containing 10%fatal bovine serum (FBS) continued researching. MTT assay and cell counting were used to detect inhibiting rates of cultured hRPE cells.Flow cytometry was used to analyse cell cycles and apoptosis.Results Treated with 1%DMSO for 48h,data of OD was significantly reduced in a dose and time dependent manner than the control groups(P<0.001).Flow cytometry detected that cell cycles varied significantly treated with 2% DMSO for 72h. The percent of the cultured cells on GI phase was increased from 41.31% to 59.65%,the percent on S and 62 phase were decreased from 32.09% to 21.21% and from 26.65% to 19.14% respectively(P<0.001),hRPE cell were arrested on GI phase.Apoptosis began to raise significantly ly at 1% level, the difference was more significant with the raising of concentration.Conclusion This study demonstrated that DMSO can inhibit proliferation of hRPE and arrest RPE cells on G1 phase induced cell apoptosis in a dose-dependent manner.
