Objective: The purposes of the experiments were to observe the effect of IL-11 on graft-versus-host disease and graft-versus-leukemia after allogenic bone marrow transplantation and related mechanism in acute lymphoblastic leukemic (ALL) mice. Methods: 20 heath SCID and BABL/C mice were divided randomly into either experimental group or control group. In experimental group mice were injected subcutaneously with IL-11 (250ug/kg) twice daily from day -2 to +7 after BMT.Mice from the control groups did not receive injection of IL-11.The model of All BMT were copied. The impact of IL-11 on CD4 and CD8T cell after transplantation was evaluated by flow cytometry; It was observed that the GVHD pathology changing of internal after transplantation in ALL mice; The survial time after transplantation was recorded; The level of tumor necrosis factor-a (TNF-a) which evidently related to GVHD was evaluated by ELISA.Results: IL-11 could decrease the quantity of CD4 T cell and increase CD8 T cell; IL-11 could increase obviously the survival time and decrease the level of TNF-a. after ALL mice transplatation; IL-11 could delay and relieve GVHD.Conclusion: IL-11 could alleviate GVHD and retain GVL effect after allogenic bone marrow transplantation.
