Objective: In this research, PDLC were obtained with tissue explants method. We studied the effects of different concentrations of BMP on PDLC's proliferation in vitro. PDLCs were cultured with β-TCP and gelfoam respectively in three dimensions. Then we discuss PDLC's attachment and proliferation, and biocompatibilities of the two materials.Methods: This experiment was divided into three parts as follows:1. PDLC culture in vitroPDLCs were derived from periodontal membrane of healthy 1st premolars that were extracted for orthodontics. PDLC cultures were established by means of tissue explants method. These cells were examined by phase-contrast microscope, scan electric microscope and acridine orange stains.2. Culture of PDLC/p-TCP in three dimensions in vitroPDLCs of 4-7 passages were seeded on p-TCP. Two weeks later, this composite were detected by phase-contrast microscope, scan electric microscope and hard tissue sections.3. Culture of PDLC/gelfoam in three dimensions in vitroPDLCs of 4-7 passages were seeded on gelfoam. Two weeks later, this composite were detected by phase-contrast microscope, scan electric microscope and acridine orange stains.Results: 1. PDLCs were long shuttle shape in vitro. Bodies of the cells were plump. Cells spreaded well and arranged in a certain orientation, looked like flame or swirl. BMP solutions with different concentrations of 20 ?200 μg/L promoted cell's proliferation obviously.2.PDLC attached to P-TCP firmly, crossed from one hole to the another and migrated into the hole. PDLCs spreaded prominences, which connected together. It showedobviously synthetic matrix.3. PDLC attached to gelfoam, spread well. Most cells were long shuttle shape. Cells grew paralleling fibers or intersected or around fibers. PDLCs spreaded prominences, which connected together to form a piece.Conclusions: 1. We could obtain plenty of, active, pure cells by means of tissue explants method.2. BMP solutions with concentrations of 20-200 g/L could enhance PDLC's proliferation.3. P-TCP could be a promising material of periodontal tissue engineering for its perfect biocompatibilities.4.Gelfoam could be used as scaffold of periodontal tissue engineering in the future for its excellent biocompatibilities towards PDLC.
