Effects of Clerodendranthus spicatus and rosmarinic acid on the glomerular Mesangium matrix and TGF-β1 Of the 5/6 nephrectomy Sprague-Dawley rats.Objective To explore the effects of C.spicatus and rosmarinic acid on the glomerular Mesangium matrix and TGF-β1 Of the 5/6 nephrectomy Spraque-Dawley rats.Methods The animal model of chronic renal failure were prepared from month 1.5~2 Sprague-Dawley rats by the way of Platt. the 5/6 nephrectomy Spraque-Dawley rats were treated with C.spicatus and rosmarinic acid for 2 months. the effects of C.spicatus and rosmarinic acid on the glomerular Mesangium matrix and TGF-β1 Of the 5/6 nephrectomy Spraque-Dawley rats were tested by immunohistochemistry method. Electron microscope was used to examine the pathology of the renal constitution of the chronic renal failure rats. meanwhile the content of creatinine , Maleic Dialdehyde(MDA) and Superoxide Dismutase(SOD) activity in the serum and of the tested animals were measured. Results 1. compared with matched control group's,the creatinine clearance rate of treated groups were higher,(P<0.05). 2. the number of fibronectin and collagen-IV in the glomerulus were significantly decreased(P<0.05).3. C.spicatus and rosmarinic acid decreased the expression of TGF-β1 in kidney (P<0.05).4.C.spicatus and rosmarinic acid had no effects on MDA and SOD in the serums of the tested rats.Conclusion C.spicatus and rosmarinic acid suppressed the expression of TGF-β1 in kidney and decreased the number of fibronectin and collagen-IV in the glomerulus, preventing glomerulus from sclerosis.so C.spicatus and rosmarinic acid might be a promising agent to treat chronic renal failure diseases.
