To investigate the role of EGFR gene and ICAM-1 gene in thecarcinogenesis and progression of human gastric carcinomas.MethodsImmunohistochemical methods were used to detect the expression of EGFRand ICAM-1 protein in 42 cases of normal gastric mucosa along with 62 casesof sporatic human gastric cancer. Meantime, we analyzed the data above withclinccopathological cancer parameters. Results(1) There was a significant difference (x2=18.97, P=0.001)in the expression of EGFR in the sporatic human gastric cancer (59. 6%) and normal tissures(16. 6%).(2) The positive stain rates of EGFR expression in the group of invasive depth T3/T4(19/26,73.1%) showed much more expression than that of T1/T2(16/36, 44.4%). And when lymph metastasis was concered, EGFR expression in the cancer with lymph metastasis(29/41,70. 7%) were higher than those without node metastasis(8/21, 38. 0%). Also, we saw that EGFR expression of the elder groups(30/43, 69. 7%) were higher than those of the younger groups(7/19, 36. 8%) in the different age groups by 60 year.(3) There were no obviously difference in the incidence of EGFR protein expression in sporatic human gastric cancer among the sexgroups, the focus size , differentiated degree, TNM stage and Lauren type groupes (P>0.05).(4) The percentage of ICAM-1 expression in cancer were higher than those in normal tissures(51.6%, 21.4%respectively) and there was significant difference (x 2=9. 552, P=0. 002) .(5) ICAM-1 expressions were distinctly different in three groups. The percentages of ICAM-1 in poorly differentiated groups (66. 6%) were higher than those in moderate, well differentiated groups(40. 0%). And ICAM-1 expressions of cancers with lymph metastasis(63.4%) showed higher than those without lymph metastasis(28. 5%). In the TNM stage groups , expressions of III/ IV stages one (65.5%)were higher than those of I / II stages groups(39. 3%).(6) There was a positive correlation between EGFR and ICAM-1 protein expression in sporatic human gastric cancer. ( r=0. 520)Conclusion(1) There exist the abnormal expressions of EGFR and ICAM-1 in gastric cancinoma. Both radios increased obviously in carcinoma tissures. There is certain correlation between the expressions of EGFR and ICAM-1 in gastric cancer.(2) The expressions of EGFR and ICAM-1 have the close relationship to the behaviour of advanced gastric carcinoma.(3) The combination detection of EGFR and ICAM-1 have the positive significance of the judge of malignant phenotype, lympg metastasis and guide to the clinical treatment.
