Purpose: Gubernaculum testis plays a major role in testicular descent and development. There are three major factors of gubernaculums testis are related to testicular descent. They are the attachment of the gubernaculum testis, active contraction and series of changes of structure and component of the gubernacum testis. Many questions are still not clear for the three parts and need to discuss. The aim of this research is to study the attachment gubernaculum testis in mice by histology observation, and observe the express of ELASTIN that reflect the series changes of gubernaculum testis and observe the express of ACTIN that reflect the active contraction of gubernaculum testis. For more def initude of the role of gubernaculums contraction and structural changes in testicular descent and development, we use several kinds of exoestrogens such as diethylstilbestrol (DES), 17-aE2 and E3 which work on the female mikes in deferent pregnant periods, and study the effect for the ACTIN and ELASTIN of the gubernaculums testis.materials and methods: Mature mice were divided into twenty-nine groups randomly, 10 every group. 1. To study the changes of gubernaculums indifferent periods, we designed ELASTIN groups and ACTIN groups, each included 7 groups, that was newborn group, born one day group, born three day group, born five day group, born seven day group, born nine day group and born eleven group.2. To study exoestrogens effect structure and component of gubernaculums testis, we designed ELASTIN group and ACTIN group too. That was 1. ELASTIN group: normal group (not given any drug), control group (given DMSO), and experimental groups were divided into DES group, 17- a E2 group and E3 group, each was given corresponding exoestrogens lOOM-g/kg. d. 2. ACTIN groups: we used random blocks design, made pair and then group randomly. 2 normal group(not given any drug), 2 Control group (given DMSO), and experimental groups were divided into early gestation group and late gestation group, and they were respectively divided into early gestation DES group, early gestation 17-aE2 group, early gestation E3 group and late gestation DES group, late gestation 17-a E2 group, late gestation E3 group, each was given corresponding exoestrogens lOOM-g/kg. d. Experimental course: firstly, female mice were placed in individual cages with male mice and the morning when a vaginal plug appeared was designated as gestation day 0 (GDO) of pregnancy. The early gestation group mice were injected subcutaneously from GDO to GD7 with exoestrogens in DMSO or with DMSO alone (controls). The late gestation group mice were injected subcutaneously from GD11 to GDIS and ELASTIN group were injected subcutaneously from GDO to GD16 with exoestrogens. Borned mice were drew the materials randomly according to scheduled time and the number of sample: they were decapitated and the low parts were fixed in 10%formaldehyde solution. Some samples were sliced serially in coronal and were stained with HE and Verhoeff's method, expression of ACTIN and PCNA in gubernaculum testis was detected by immunohistochemical methods. Histological changes were observed with light microscope.Results: The gubernaculum testis attached to the epididymis and testis, and scattered bundles inserted in the testis, binding to testis tightly; the distal of the gubernaculums attached to pubic symphysis, a part of it attached to Abdominal wall; gubernaculum testis consisted of the proximal gubernaculum cord and distal gubernaculum buld. Elastic fibers of Gubernaculum in born seven day group increased than that of newborn group. In the same way, ELASTIN of gubernaculum in born seven group, born nine group and born eleven group increased than newborn group. ACTIN of gubernaculum in born seven group, born nine group and born eleven group increased than newborn group. ACTIN of gubernaculum in newborn mice that have been exposed to exoestrogens during early gestation decreased than that of normal group and also decreased than that during late gestation.
