Objective: To observe the anti-tumor action of Yanghe decoction, and exploring its possible local immune mechanism, and to present a experimental evidence for warm-method treating tumor.Methods: 1 Experiments on the Anti-tumor Effects of Yanghe Decoction. Mice were inoculated with S180(A) and HepA tumor cells in lateral subcutaneous tissue of right hind legs, respectively. The tumor-transplanted mice were divided into five groups by random: low dose of Yanghe decoction group, middle dose of Yanghe decoction group, high dose of Yanghe decoction group,chemotherapy group and model control group. 10 days after drug administration, the tumor inhibitory rate was calculated from the tumor weight of each group. Mice were inoculated with EAC tumor cells in abdomen. Dividing groups as same as above. 9 days after drug administration, the lifespans of tumor-bearing animals were observed and the life-prolonging rates were calculated.2, Experiments on the Effects of Yanghe Decoction on IL-2, IFN-r, TGF-B1 and the Cytotoxicity Activitity of Tumor Infiltrating Lymphocytes(TIL) to Auto Tumor Cells(ATC) in Ascites of EAC-transplanted Mice. Mice were inoculated with EAC cells in abdomen. The tumor-transplanted mice were divided into five groups by random: low dose of Yanghe decoction group, middle dose of Yanghe decoction group, high dose of Yanghe decoction group,chemotherapy group and model control group. 9 days after drugadministration, Ascites of mice were abstracted and the density of IL-2. IFN-r, TGF- B1 was determined by ELISA method.,and the cytotocity activitity of TIL to ATC was determined by MTT method.Results: 1, The tumor-inhibitory rates of low. middle and high dose of Yanghe decoction groups to S180(A) were 0.67%, 18.79%, 39.60%, and the tumor-inhibitory rates to HepA were 2.72%, 19.05%, 34.69%, and the weight of tumor of high dose of Yanghe decoction group was lower than model control group, the defference between two groups has a statistic significance(P<0.01). The lifespan-prolonging rates of low, middle and high dose of Yanghe decoction were 26.56%, 48.44%, 36.72%, and the lifespans of middle and high dose of Yanghe decoction were longer than that of model control group (P<0.01),and the lifespan of low dose of Yanghe decoction was also longer than that of model control group(P<0.05). 2, IL-2 of low, middle and high dose of Yanghe decoction groups were 76.75 + 22.47pg/ml, 97.97 + 24.23pg/ml, 112.11 + 24.06pg/ml, IFN-r of low, middle and high dose of Yang he decoction groups were 13.88 +4.26pg/ml, 16.93 +4.24pg/ml, 17.15 +4.78pg/ml, IL-2 , IFN-Y of middle and high dose of Yang he decoction groups were higher than that of model control groups, and the differences have statistic significance (P<0.05).TGF- B1 of low , middle and high dose of Yanghe decoction groups were 3906.25 + 1414.90pg/ml , 2695.31 + 1610.32pg/ml , 2304.66 + 1392.92pg/ml, TGF-B1 of middle and high dose of Yanghe decoction groups were lower than that of model control groups, and the differences have statistic significance(P<0.05). The cytotocity activity of TIL to ATC of low, middle and high dose of Yanghe decoction groups was 9.52% + 3.49%, 12.93% + 3.73%, 14.14% + 2.93%. The cytotocity activity of TIL to ATC of middle and high dose of Yanghe decoction groups was higher than that of model control group. The defferences have statistic significance(P<0.05).Conclusions: 1 High dose of Yanghe decoction has the anti-tumor action on S180(A) and HepA, and low , middle and high dose of Yanghe decoction group canprolong lifespans of EAC-bearing mice. 2 Middle and high dose of Yanghe decoction can increase the density of IL-2. IFN-r and decrease the density of TGF- B1. Middle and high dose of Yanghe decoction can activated the cytotoxicity function of TIL to ATC in vivo in EAC. Therefore, Adjusting the local immune of tumor is a possible mechanism of the anti-tumor action of Yanghe decoction.
