ObjectiveThe research is on the context of psychosomatic medicine that to discover the mechanisms and influence of emotion stimulation on neuroendocrine function.behavior when DM and the regulatory function of composite formula Xinshen NO.5 which has the efficiency of regulating Qi lowering rebellious qi and dispersing accumulation.MethodThe rats models of DM were made by cauda vein injection with little dosage of STZ ,cooperated with adipose diet at the same time. Then, exert emotion stress (irregular photoelectricity) to rats to observe the influence to it. The experiment were composed of two parts: acute stress and chronic stress ,each part were divided into five groups at random :control group^ stress group > STZ group v stress+STZ group and treatment group by Chinese herbs. Use immunology of radioimmunity, immunohistochemistry, RT-PCR technology examined the chemical changes of blood suga^ insulin^glycago^neuropeptide Y^ypothalamus-pituitary-adrenal axis under emotion stimulation ;And to evaluate the influence of emotional stimulation to rats 'action by the open-field test, in synchronism to observe the regulatory functions of decoction.Result1. STZ cooperated with adipose diet can elevate rats' blood sugar level(average:25.3mmol/L), destroy P matrix ,which can result in DM rat model.2. Emotional stimulztion can elevate rats' blood sugar > glycagon, reduce insulin. Index of the treated group were ameliorated.3. The emotional stimulation can increase the transcription level of c-fosmRNA and the translation level of FOS protein and the serum cortisol level (P<0.05).The influence to diabetes rats were weaker than to control rats. Compound decoction can alleviate the influence.4. The transcription level of NPYmRNA and the translation level of NPY protein potentiate under the condition of diabetes . Emotional stimulation can increase the transcription level of NPYmRNA of control rats. The condition of diabetes rat was opposite. Chinese traditional medicine can regulate the change of diabetes rat.5. DM rats had contrast to control rats. Emotional stimulation can result to abnormal action to both groups(PO.01),the treated group changes was light.Conclusion1. The animal model was made by STZ cooperated with adipose diet. And emotional stimulation can make rats' blood sugar elevated ; secrete of glycagon and cortical increased; insulin reduced,macroscopical behavior changed. That indicated emotional stimulation can aggravate disturbance of carbohydrate metabolism and which like the DM model of Bing and Zheng compounded .2. Emotional stimulztion can aggravate diabetes rats' disturbance of carbohydrate metabolism perhaps has the relationship with hypothalamus-pituitary-adrenal axis and sympathetic- adrenal axis.That reflects Qi disorder maybe the resulats of lost of coordination between mentality and physiology.3.The responsibility of diabetes rats are weaken. Emotional stimulation can result to abnormal action, which behaved as excitement chiefly, but can' transformed to depress with the process of the stress . Macroscopical behavior of rats reflect the disorder of Qi.4.Xinshen No.5 may interfere the activation of hypothalamus-pituitary-adrenal axis and sympathetic nerve to lighten abnormal influence of emotional stimulation on diabetes rats, and which can regulate the macroscopical behavior of rats simultaniely.
