Effect Of Smoking On The Expression Of PDGF In The Rats' Cerebral Vascular Endothelial Cells

The ischemic cerebrovascular disease (ICVD) has been commonly seen in the disorder of nervous system. The incidence and the mortality of the disease have been keeping high, and it usually leaves the survivors with various dysfunctions. Epidemiology study demonstrated that there were eminent relation between smoking and ICVD.According to document, lesion of the endothelial cell is the primary cause and first-emerged pathologic change of ICVD, and smoking is one of important but avoidable risk factor. This article aimed at building a possible bridge between smoking and ICVD via observing the expression of PDGF-A and PDGF-B in rats' cerebral vascular endothelial cells, clarificating the ICVD mechanism caused by smoking.Material and MethodFifty male wistar rats were randomly groupered into five groups : long-term-massive cigarette smoking group short-term-massive gronp, long-term-morsel group, anti-smoking group, normal control group. Each group has 10 wistar rats. The expression of PDGF-A and PDGF-B was localized using immunohisto-chemistry technique, The positive cells was calculated by microscope, the data was expression by X ?S and was analyzed using t test.ResultThe positive cell of PDGF-A and PDGF-B showed hrown and located onlyin the endothelial cell. There was a small quantity of expression of PDGF-A and there was not a significant difference among all the groups. The expression of PDGF-B was expressed in all wistar rats'cerebral vascular cells, but seen more those under cigarette smoking. Contrast to the normal control group, anti-smoking group is not significant ( P > 0. 05 ) short-term-massive group , Long-term-morsel group and long-term-massive is significant increment(p <0.01).DiscussionThe structure and function's disability of endothelial cells play important rolls in the occurrence and progress during cerebral disease. Those risk factors that cause atherosclerosis at the same time case EC functional lesion. At present , People think that EC lesion is an early incident of atherosclerosis.At normal condition EC excretive few PDGF. Under pathologic circumstances , PDGF is expressed widely and it is an early incident of PDGF on EC in the cerebral thrombus.PDGF secreted by the endothelial cell is composed of two chains, A and B. PDGF have multifold functions. Among these, chemotaxis and vasoconstrictor pep tide are important.Our results showed that the expression of PDGF-B was expressed in all Wistar rats'cerebral vascular endothelial cells but seen more in those under cigarette smoking, long-term group is significant among groups and the following are short-term-massive group, long-term-morsel group, anti-smoking group and normal control group. Short-term-massive cigarette smoking group contrast to normal control group, the expression is significant, it show that smoking can cause PDGF-B elevated after short term. Long term-massive cigarette smoking group contrast to short-term-massive cigarette smoking group, the expression is significant, it show that there are time effect relationship between cigarette smoking and the expression of PDGF-B. Long-term-massive cigarette smoking group contrast to long-term-morsel cigarette smoking group, the expression is significant, it show that there are dose effect relationship between cigarette smoking and the expression of PDGF-B. The expression of PDGF-B was decreased in rats'cere-bral vascular endothelial cells after antismoking and statistical analysis shows there is no significance versus control group. There was a small quantity of expression of PDGF-A and there was not a significant difference among five groups. It is show that PDGF-A is not significant during cigarette smoking case the lesion of the endothelial cell. Cigarette smoking cause EC express much PDGF-B, it cause blood platelet adhesived to EC and accelerate SMC move to end- omembrane.
