Cadmium (Cd) is an important industrial and environmental pollutant, through food chain and drinking water, that causes sever damage to a variety of organs including the lung, liver, kidney, testis and placenta. The kidney is the target organ for chronic Cd poisoning .There is evidence that long-term exposure to Cd leads to renal tubule dysfunction. It was reported that cadmium chloride was found to induce necrotic in renal cells of nephrotoxicity studies. Recently, some people thought that apoptosis was one of reasons in Cd- nephrotoxicity abroad. But few studies has not been reported that Cd produced apoptosis in renal cells in China. Furthermore, the molecular mechanism of apoptosis- cadmium induced for renal cells has not been clear. In the present study, the apoptosis Cd-induced for LLC-PK] cells was studied to clarify the molecular mechanisms of renal lesion induced by cadmium.PART ONEThe effect of proliferation in LLC-PK1 cells induced by cadmium with or without serumTo identify the effect of porcine renal proximal tubule epithelial cells LLC-PK) induced by cadmium in medium with or without serum. Cellviability was measured by MTT system and the morphological changes were examined using Image-Pro Plus software. The results showed: Incubation with equal concentration of CdCl2 for 8h and 12h,the viability of LLC-PK1 cells with serum in medium was higher than that of without serum. Without serum group, LLC-PK1 cells with 40umol/LCdCl2 for 12h in medium, most cells were floated; But with serum group, a few of cells floated at the same time and concentration. Under the equal concentration of cadmium, the morphological changes exhibited that the injury of cells without serum was more serious than those of with serum. The results indicated that serum could inhibit the cytotoxicity cadmium-induced in LLC-PK1 cells, and suggested the study for cytotoxicity cadmium- induced in LLC-PK1 cells should be measured without serum in medium to decrease the effect of serum.
