Objective: The bladder urodynamic manifestations of alloxan-induced diabetes mellitus (DM) rats were investigated. We tried to find a relationship of urodynamic and detrusor histopathologic abnormities in the alloxan-induced diabetic rats and to illuminate the mechanism of diabetic patients' bladder dysfunction.Methods: The diabetic rats were induced by injection alloxan. The rats' bladder functions were evaluated. The detrusor histopathologic structure was observed by optional and electric microscopy.Results: The experiment was comprised 10 control rats, 10 rats of six weeks after induction DM and 10 of ten weeks after induction. 1. All the control rats' bladders were able to contract, void and empty the bladder. One rat's bladder (1/10) was acontractile in 6 weeks group and two (2/10) in 10 week group. 2. The maximum detrusor pressure (Pdet,max) in control group and two experiment group was alike (p>0.05). The DM rats' bladder volume and residual urine volume were higher than control rats' (p>0.05). 3. To observed the HE, Van-Gieson's and Masson's stained sections with optional microscopy, the muscle bundle ranged regularly, compacted in control rats. In the DM rats, the muscle bundle ranged confusedly, collagen among muscle bundle reduced compared to control rats. The changes were more severe in six weeks group than ten weeks group. 4. In contrast to the control rats, the diameter of smooth muscle cell (SMC) increased. The gap between two detrusor cells decreased. Collagen fiber density decreased among detrusor cells in muscle fascicles. The density of dense bands decreased. The quantity of dense patch and intermediate junction decreased.Conclusions: Bladder dysfunctions were found in early stage of alloxan-induced6diabetic rats. Thus, it's essential to perform urodynamic study in diabetic patients to identify their bladder functions.
