Protect Effect And Mechanism Of VLC On Myocardial Ischemia

In this paper,protective effect of VLC on myocardial ischemia was investigated.After establishing a hypoxia-reoxygenation model on neonatal rat myocytes,we investigated the machanism of its protective effect. l.The establishment of acute myocardial ischemia model on dogsFrom the literature,two methods were used in establishing myocardial ischemia models: (1) the left anterior descending artery (LAD) occlusion was employed proximal to the first diagonal branch;(2)Left anterior descending artery(LAD) occlusion were employed under the middle point.In the above method,the ischemia area was large and the motality was high.Therefore,the left marginal branch and diagonal branch occlusion were employed in our method.Conclusion: the method we used was much better. 2.The protective effect of VLC on acute myocardial ischemia on dogsEstablish an acute myocardial ischemia model on dogs,then left marginal branch and diagonal branch of left anterior descending artery(LAD) occlusion were employed in this study.The total elevating millivolts of ST (Σ ST) ,the leads number that ST elevating over 2 millivolts (NST) ,the number of pathological Q waves(NQ) on epicardial ECG;the contents of LDH, CPK in serum and the infarction index were observed to evaluate the protective effect of VLC;at the same time,heart rate and blood pressure were measured.Compared with control group,VLC decreased the ΣST, NST, NQ values on epicardial ECG, GPT, AKP content in serum .Simultaneously,the high dose group could decrease heart rate after ischemia.lt suggested that VLC had protective effect on myocardial ischemia. 3.The mechanism research of VLC on acute myocardial ischemiaSerum pharmacology method were used in this study.After 5 days treatment with VLC 32mg/Kg , 16mg/Kg ig. in rats,the serum were separated and added into culture medium.Then,the myocardial cells of neonatal rats were cultured under hypoxia condition for 24h,followed by reoxygenation for 4 h.Finally ,the LDH and MDA were examined.To get the apoptosis rate,the FCM were used.VLC has anti-apoptosis effect in myocardial cells by decreasing the production of ROS.
