Objective : It has been reported that aldosterone can be synthesized in extra adrenal tissues including liver.aldosterone is a strong stimulator of fibrogenesis and mitogenesis which exerts a marked effect on the proliferation of myofibroblasts and the production of collagen, therefore,many people have realized that aldosterone may play an important role in the progression of hepatic fibrosis. Some experiments show that inhibitor of aldosterone contains the function of preventing and interrupting hepatic fibrosis in overseas and china. Further evidence finds that antisterone (an inhibitor of aldosterone) has a fibrogenesis inhibiting effect before the middle stage by inhibiting the binding of aldosterone to its receptor. At the late stage,hepatic fibrosis becomes irreversible because of the parti- cipation of many factors. YiGanXian,as a complex prescription of Chinese material medica ,can promote blood circulation to dissipate blood stasis, soften hard lumps and dispel nodes ect. It can prevent and reverse the progress of hepatic fibrosis ,this is identified by the previous study of our department.One purpose of this study is to observe the effect of antisterone, antisterone and YiGanXian on preventing and interrupting hepatic fibrosis and wether YiGanXian can reparative the inadequacy of antisterone on the late stage;The other is to conjecture the mechanisms of aldosterone accelerating liver fibrosis and YiGanXian preventing hepatic fibrosis.Methods: the study selected male Wistar rats 60,who were divided into two groups randomly at the beginning .In the model group,the rats were injected with 40% carbon tetra- chloride; In the antisterone group, the rats were injected with 40% carbon tetrachloride, antisterone was given intragas- trically.when the sixth week was over, the antisterone group were divided into 2 groups ,one group was given antisterone as before; the other group was given YIGanXian 5 weeks. After 3,6,9 and 12 weeks, animals were sacrificed , Masson and HE hisochemical staining of liver tissuse samples were Performed; the area of collagen ,liver homogenate hydropropline (hyp),the activate HSC cells who contain α-SMA were examined; quantitative for unitarea areadensity integral lumi- nosity PAI-1 immunohistochemistry of liver tissue samples was performed .Results: ①In model group, before 9 weeks , the grade of fibrosis ,the area of collagen and the homogenate hyp increased gradually, at 12 weeks, having stopped carbon tetrachloride treatment 3 weeks, the liver damage had recoured, but the grade of fibrosis,the area of collagen and the homogenate hyp in the model group didn't decrease compared with 9 weeks' model group.②In the antisterone group,the grade of fibrosis ,the area of collagen and the homogenate hyp were less than those in model group before 6 weeks, at 9 weeks , there was no significant difference between these two group. This confirmed that antisterone could really preventing the development of liver fibrosis,but antisterone had its limitations, it had no significant effects at the late stage.③In antisterone and YiGanXian group, at 9 weeks ,the grade of fibrosis ,the area of collagen and the homogenate hyp were less than model group, this indicated that YiGanXian could prevent the development of liver fibrosis.At 12 weeks,the grade of fibrosis,the area of collagen and the homogenate hyp in antisterone and YiGanXian group were lower than 9 week's antisterone and YiGanXian group,we could say that YiGanXian could reverse the liver fibrosis.④Before 9 weeks,the level of α-SMA and PAI-1 went up gradually in both model group and antisterone group ,but antisterone group was lower, it could be conjectured that the mechanism of aldosterone accelerating hepatic fibrosis was increasing the level ofα-SMA and increasing the expression PAI-1.⑤At 9 weeks,In antisterone and YiGanXian group,the level ofα-SMA and the expression of PAI-1 were also lower than the model group,we guessed the m...
