| Objective: To investigate antitumor effect of biaminotetradecanoic acid (D-82) in vivo & vitro, and to explore the mechanisms of antitumor effects.Methods: The cytotoxic effects of D-82 on various tumor cell lines cultured in vitro were determined with trypan blue dye exclusion test, MTT method and growth curve method. At a tolerable dose level, D-82 was administrated to treat mice transplanted solid tumors S180 and U14, tumor growth inhibition was observed. Apoptosis morphological transformation of HL 60 cell induced by D-82 was detected by the phase contrast microscope,Giemsa staining and AO-EB staining. Alteration of cell cycle was analyzed by flow cytometer. DNA ladder was detected with agarose gel electrophoresis.Results: D-82 induced significant growth arrest in various malignant tumor cell lines cultured in vitro and effectively inhibited in-vivo growth of mice implanted solid tumor of U14 and S180. Following 3μg/mL and 6μg/mL D-82 treatment for 24h and 48h, flow cytometry cell cycle analysis revealed the increasing presence of sub-G1 apoptotic peaks. After treatment with D-82, agarose gel electrophoresis could demonstrate DNA ladder, typical of apoptosis.Conclusion: D-82 had obvious antitumor activity in vivo & vitro. D-82 could induce the apoptosis of HL 60 cell. |
