OBJECTIVE To investigate the efficacy of combined use of bone morphogenetic protein(BMP) and vascular endothelial growth factor(VEGF) in repairing segmental bony defects. METHODS According to the plan, BMP was mixed with 0.1mol/L PBS(pH=7.4) and then homogenized into suspension, VEGF and polyvinylpyrrolidone(PVP) were mixed with distilled water and then made into solution with viscosity. The suspension and solution were mixed thoroughly with the vituline cancellous bone as the vector under vacuum and lyophilized. Then receive three kinds of composite: vituline cancellous bone carrier/BMP+PVP/VEGF,vituline cancellous bone carrier/PVP/BMP,vituline cancellous bone carrier/PVP/VEGF. Next, implant different kinds of composite and vituline cancellous bone+PVP carrier in the 15-mm-long continuous defects in radius of adult rabbits. The animals were sacrificed 3,8 and 12 weeks after operation, the bone repair process was investigated using physically, roentgenogram, histological observation and biology mechanics detection. RESULTS The vascularization in vituline cancellous bone carrier/BMP+PVP/VEGF or vituline cancellous bone carrier/PVP/VEGF transplanted applied areas were better than those in vituline cancellous bone carrier/PVP/BMP,vituline cancellous bone+PVP carrier 3,8,12 weeks after operation. Histological and roentgenogram examination showed that vituline cancellous bone carrier/BMP+PVP/VEGF composite was implanted were repaired more rapidly than those other three groups. Biologic mechanics showed that on the vituline cancellous bone carrier/BMP+PVP/VEGF composite side the callus was growing more quickly and vigorously, the bone density was higher and the antibending force was stronger than those other three groups after 12 weeks. CONCLUSION The composite of vituline cancellous bone carrier/BMP+PVP/VEGF can efficiently repair bony defects, which can improve not only the vascularization but also the differentiation of primitive cell, and the capability of vituline cancellous bone carrier/BMP+PVP/VEGF is the best of all by the way of satisfactory osteoconduction, osteoinduction. Vituline cancellous bone carrier/BMP+PVP/VEGF composite may be feasible for diaphyseal defect restoration, and the combination of BMP and VEGF is synergic in inducing bone formation.
