Objective:Positively and specificly immunological oncotherapy is a therapy that using kinds of tumor vaccine(TV) to induce anti-tumor effects of organism ,which is a important method to prevent the recurrence and transition of tumor.because of neoplastic load and immunological tilt of neoplastic host,the effects of therapy are not very good.we want to find out a kind of immunomodulator which can intensify the remedial effect of tumor vaccine.CpG motifs is a kind of oligodeoxynuleotieds(ODN) with non-methylcytosine and non-methylpurine as it's center. Studies show that CpG ODN can activate the immunologic competence of organism, especially cellur immunity ,and can be used as a kind of immuno-adjuvant,we have studied differently the effect of tumor and CpG ODN in our laboratory and proved the manifest anti-tummor effect of TVPBH which could be used in clinic. At the same time,we have proved the manifest tumor-suppressed effect of CpG ODN,which can active the the cellular immunity of mice but have on direct cytotoxicity to tumor cells.on this bases,anti-tumor effects induced by hyperthermal solid tumor vaccine(HSTV) associated with CpG ODN were explored in this expriment in order to provide the evidence for application of HSTV and CpG ODN to treat the patiend of tumor. Methods:32 BALB/c mice were borne tith tumor and divided into 4 groups at random:NS control group,tumor vaccine treatment group,CpG ODN treatment group, tumor vaccine and CpG ODN co-used treatment group .from the 2th day after bearing tumor,NS(0.1ml),tumor vaccine(2×107/0.1ml per mouse), CpG ODN(10μg/0.1ml per mouse), tumor vaccine+ CpG ODN({tumor vaccine(2×107)+ CpG ODN (10μg )}/ml per mouse) was respectively injected into the four groups of mice by subcutaneous(SC),every other day,five time totally.the four groups of mice were killed after 15 day,then, tumor were taken off,they were weighed firstly for mesurement of inhibition rate .at the sime time, spleen and abdmental extravasate were collected to detect NK,CTL,MФcells' kill and wound activity,serum was colleceted to detecte IL-12,IL-10.Results:1.Tumor inhibited rate of mouse used as a subject of expriment in different groups:tumor vaccine,CpG ODN or co-use of tumor vaccine and CpG ODN can all suppress the growth of tumor.tumor inhibited rate of respective groups is 48.92%,75.0%,86.67%.contrasting with group of NS ,there is obvious diffrence in tumor inhibited rate(p<0.05). contrasting with group of tumor vaccine or group of CpG ODN, there is obvious diffrence in tumor inhibited rate in group of co-use of tumor vaccine and CpG ODN(p<0.05).2.NK cell's kill and wound activity of mouse used as a subject of expriment in different groups: contrasting with group of NS, there is no obvious enhance in NK cell's kill and wound activity in group of tumor vaccine(p>0.05),but there is obvious enhance in NK cell's kill and wound activity in group of CpG ODN(p<0.05). contrasting with group of tumor vaccine, there is obvious diffrence in NK cell's kill and wound activity in group of co-use of tumor vaccine and CpG ODN (p<0.05). contrasting with group of CpG ODN, there is no obvious diffrence in NK cell's kill and wound activity in group of co-use of tumor vaccine and CpG ODN(p>0.05). 3.CTL cell's kill and wound activity of mouse used as a subject of expriment in different groups: contrasting with group of NS, there is obvious enhance in CTL cell's kill and wound activity in group of tumor vaccine or group of CpG ODN(p<0.05). contrasting with group of tumor vaccine or group of CpG ODN,there is obvious diffrence in CTL cell's kill and wound activity in group of co-use of tumor vaccine and CpG ODN(p<0.05).4.MФ cell's kill and wound activity of mouse used as a subject of expriment in different groups: contrasting with group of NS, there is obvious enhance in MФ cell's kill and wound activity in group of tumor vaccine and group of CpG ODN(p<0.05). contrasting with group of tumor vaccine, there is obvious diffrence in MФ cell's kill and wound activity in group o...
