Experimental Research On The Growth Inhibit Effect Of Thermochemotherapy For Liver Cancer Cell Line SMMC-7721

OBJECTIVE:Studying inhibit effect,cell cycle changes and possible effect mechanism to the liver cancer cell SMMC-7721 line.Observing the endure situations that normal pro-generation liver cell human to themochemotherapy .METHODES: Separating and cultivating human pro-generation liver cell in vitro by collagen enzyme methode;Using the themochemotherapy dosage that 43℃,30min+CDDP1.0ug/ml,43℃,60min+CDDP1.0ug/ml,43℃,90min+CDDP1.0ug/ml to dispose human pro-generation liver cell in 24,48,72 hours, Observating the changes of cell morphology in light microscope,and dectecting clture medium ALT in 24,48, 72 hours after disposing,in the end,get a safe therapy dosage ,then using the dosage to treat liver cancer cell SMMC-7721.Observating the changes of cell morphology and ultrastructure in light and electron microscope,and using MTT methode to examine the living degree of liver cancer cells that were disposed. Flow cytometry detects cell cycle.RESULTS:(1) The growth inhibit effect of the human pro-generation liver cell in 43℃,30min+CDDP 1.0ug/ml,43℃,60min+CDDP 1.0ug/ml,the quantity of ALT increased temporarily;however,in 43℃,90min+CDDP1.0ug/ml,results indicated that Human pro-generation liver cell destroyed obviously and the quantity of ALT increased continuously.(2) Treating liver cancer cell line SMMC-7721 in 43℃,60min+CDDP 1.0ug/ml, light microscope ,electron microscope,and MTT results indicated that the dosage can inhibit liver cancer cell line growth,and the inhibit degree of thermochemotherapy group is larger than hyperthermia group and chemotherapy group.(3)Flow cytometry detects the G1 stage cell are remarkably increased and S stage cells decreased in thermochemotherapy group.CONCLUSIONS:(1) There is no obviously inhibit effect to human pro-generation liver cell in 43℃,60 min+CDDP 1.0ug/ml therapy dosage ;but the dosage can obviously inhibit the growth of the liver cancer cell line SMMC-7721;(2) In 43℃,60 min+CDDP 1.0ug/ml therapy dosage, the inhibit degree of thermochemotherapy group is larger than hyperthermia group and chemotherapy group.(3)Continuosly thermochemotherapy disposeing is more obviouse inhibit effect than once ;(4)Thermochemotherapy inhibit liver cancer cell line by changing cell cycle;(5)Thermochemotherapy possibly stares up apoptosis program to inhibit liver cancer cell growth.
