Objective: To study the correlation of MRI features with tumor angiogenesis, proliferation cell nuclear antigen(PCNA) expression and tumor malignant degree in astrocytomas.Methods: Forty cases of astrocytomas proved by surgery and pathology were examined with MRI scanning before operation,monoclonal antibody FVIII and PCNA were expressed through immunohist chemical staining by means of SP metheds. Tumor micro vessel density(MVD) was examined by searching for the best stained area under low power lens, and then change into high power lens to select three different visual fields, and calculate the average. The expression of PCNA was examined by searching for 1000 cells under high power lens on many visual fields at random to calculate the percents of PCNA positive labeling cells. The SIEMENS 0. 2T common conduct magnetom open viva was used, transversal ant sagittal orientation have been scanned as routine methods and coronal scan was performed in partial cases. SE T, weighted imaging (TR/TE=420. 0/15. 0ms) and SE T3 weighted imaging (TR/TE=6000. 0~6285. 0/134.0ms) were obtained and contrast enhancement imaging obtained by injecting (d-DTPA(0.1mmol/kg) through cubital vein. Tumor size, range of perituiroral edema, tumor contrast enhancement degree and tumor margin shapes were measured with MRI. Results: 1. Tumor MVD was closely positively correlated with PCNA labeled index(LI)(r=0. 905 P < 0.001); 2. There were significantly different between the different grade of astrocytoma on MVD and PCNA-LI(P< 0.001);3. The extent of peritumor edema siowed by MRI was closelycorrelated with MVD and PCNA-Ll,and they were significantly different between the different peritumoral edema grades (P < 0. 001) ; 4. MVD and PCNA-LI were no significantly different between the different size of tumor (P>0. 005) ; 5. MVD and PCNA-LI were significantly different between these tumor with good margin shapes or bad margin shapes(P<0. 001) ;6. MVD and PCNA-LI were significantly different between different contrast enhancement degrees (P<0. 001).Conclusion: The MVD and the PCNA expression was certainly correlative with MRI appearance. MRI features could indirectly reflect the biological behavior of astrocytoma as well as the grade of micro vessel hyperplasia and the proliferative activities of astrocytcmas. Therefor MRI features play an important role in assessing the malignant biological behavior of astrocytomas.
