Expression Of BMP-2 With Combined Use Of Spongy Bone Particle And Bone Marrow In Repairing Long Bone Defects

Objective:To study the mechanism of combined use of mucle , spongy bone particle and bone marrow (compound periosteum particle )in repairing long defects by observation of expression of BMP-2: Methods.24.adult, male, or female, weigh from 2. okg to 2.5kg. new Zealand rabbits were randomly divided into 6 groups. Every group being 4 rabbits, pick up left of rabbits as test groups, and the right of rabbits is control group.Disinfect as usually operation. 20mm standard bone defect modal was procedured bilaterally in the middle radial shaft of rabbit with bone grinding machine, shut bone marrow socket with bone wax. taking spongy bone of ilium making particle mixing with red bone marrow from ilium repair left radical bone defect. The right radical defect is left and make it growth randomly as control group, the rabbit inject penicillin by 100ku/kg three days after operation in order to prevent wound infection. Then Sacri f iced at 3 days. 1.2.3.4.6.8 weeks after surgery for histological observation immunity chemical staining of BMP-2 and in situ, hybridization of theircDNA probes. Results: (l)Histologically observation: after repairing the long bone defect with spongy bone marrow, it pass inflammation reaction.. ossification and turn into normal bone tissue gradually. (2) Two groups are inflammation reaction when test group and control one operation for 3 days, at the same time BMP-2 is expression of negative reaction. Expression of BMP-2 is expression of weak positive when they were operated for weeks, it is expression of stronger positive when they were operated for 4 weeks, it is expression of positive when they were operated for 2,6,8 weeks. However, BMP~2 of expression is expression of negative reaction in the control groups when they were operated for 1 weeks and it was positive when they were operated for 1 weeks and it is weaker expression when operation for 2,6,8 weeks. Conclusion: (l) BMP -2 is an important factors in repairing the hone defect. (2) Genarally. BMP expression can speak that this operat ion way is effect ive , in spongy bone combined tise of spongy bone and bone marrow repairing bone defect . it can reduce the time of repairing bone defect and give some theory reliance to clinic...
