| A systematic isolation , purification and pharmacological research ofpolysaccharide of Mytilus Coruscus from Shengsi maritime space in ZhejiangProvince of China were performed by means of active detection of cell-proliferationreaction model of T and B Lymphocyte, bioactivity-oriented isolation and theDEAE-cellulose, Sephadex G-100, HPGFC etc. 9 polysaccharide compounds:MA(112,739), MD1(117,761), MD2(15,417), ME1(121,136), ME2(10,174),MF4(113,653), MF5(67,938), MF6(39,100) and MF8(12,554) were first obtainedfrom its aqueous extracts. The physical-chemistry property of these ninepolysaccharide from Mytilus Coruscus were analysised in accordance with the variousspectroscopy techniques (UV,IR,GC/MS,1HNMR) and chemical methods (Hydrolysis,Acetylation,periodate oxidation, Smith degradation, Methylation, Sulfate-Carbazolereaction,etc.). All these 9 polysaccharide compounds were heteropolysaccharides,which had considerably branching chains and extremely complicated composition,containing fucose, glucose, mannose, galactose, amino sugar, and glycuronic acidbasically.. Some of them also contain rhamnose, arabinose and xylose. Thepolysaccharide compounds (MA,MD1,ME1,ME2,MF5,MF6,MF8) were identifiedas α -configuration and the others (MD2 , MF4) as β -configuration. Theirconfigurations of glycosidic bonds were mostly 1,4-glycosidic bonds, and 1,6-glycosidic bonds oriented mainly at the bifurcate of branching. Their pentoses wereall connected with1,3-glycosidic bonds. The intermolecular deoxyhexose mainlylocated in the terminal group of the polysaccharides. The gained polysaccharides hada quite different molecular weight, different proportion of the content ofmonosaccharides in polysaccharides possessing the same components and differenttypes and contents of glycosidic bonds in some saccharides. All these 9 polysaccharide compounds could promote cell-proliferation of T and BLymphocyte. But MD1, ME1, MF4 and MF5 could significantly promotecell-proliferation of T and B Lymphocyte. MF4 could inhibit remarkably the growthof hypodermic implanted mice Lewis lung cancer sarcoma in vivo. In this subject, Our studies focused on water-extract polysaccharide of MytilusCoruscus and its pharmacological effect, we have systematically engaged in anin-depth research and discovered manifold bioactive macro-molecular polysaccharidecompounds which could be exploited as novel preparations for anti-tumor andimmunological enhancement preparation. Combining with the cytotoxic anti-tumordrugs, the compounds can enhance the immunity and anti-tumor effect. Scientificfoundation has been provided for development and research of immunologicalenhancement preparations which is quite significant in developing and utilizingabundant marine biological resources of our country. The research has enrichedcontent of marine pharmacology and phytochemistry, which could be used asreferrences of the study on marine bioactive constituents and pharmacologicalfunction. As a new anticancer drug, polysaccharide MF4 is in pre-clinic study. We arein the process of applying for patent of the for the polysaccharides. |
