Background and purpose:The lung cancer is one of highest malignant tumor in the morbidit and mortality rate .having no the valid method to diagnosis earlier so far. Thanks for technical of gene chips perfectly,it is possible to analysis large-scale lung cancer gene expression , looking for lung-cancer-related gene. However, it is not discovered that the accurate lung-cancer-related gene by now. Aims: 1.Establishing the database of lung-caneer-related gene according to the different pathologic type.2.Looking for the biomarker gene of lung adenocarcinoma, providing the theories for molecul pathology diagnosis and treatment clinical tumor prognosi and the choice of the treatment of the lung cancer.Material and method:We make use of the gene chips containing 128000 cDNAs to screen gene expression profile in lung cancer tissues for finding out the obvious adjusting up or descending genes, then by the creature information analysis, Northern blots analysis will be used to verify the related gene in tumor. Analyze the target genes expression in tumor tissues, search the cancerous and high expression gene in lung adneocarcinoma (biomarking gene), 1. Firstly, the 10 lung carcinoma patient specimens used in this study were provided by Changhai Hospital at the Second Military Medical University from September 2003 to March 2004 (shanghai,china). The cancerous lung tissues and para-neoplastic tissues(beyond 6cm) were excised from patients post mortem and placed immediately into liquid nitrogen for RNA and protein preservation. Samples of human lung cancer contain the three major subtypes [squamous cell carcinoma (SCC), adenocarcinoma and small cell lung cancer]. Then,they were detected by cDNA microarray (12800chip).According to Ratio>2 orRatio0. 5 ( CY5/ CY3) ,we picked out the numerous genes related with lung cancer , and those genes were shown in tables according the pathologic type of lung cancer2. The verification of the related gene in lung adenocarcinoma:( Northern blot analysis). Because analysis research of the gene expression of all pathologic types lung cancer is a huge and insurmountable work, and the limit with time and money, we chosen the related gene in lung adenocarcinoma as the target gene for Northern blot analysis. We screened related genes in lung adenocarcinoma again, 7 differences gene were chosen as target genes for Northern blot analysis, according the criterion of genes identified with differently expressed. (Ratio>4 or Ratio<0.25) to study the mRNA complete of genes and the expression in the tumor. As a result we discovered for the very first time that Metallothionein-3 gene ( MT-3) is highly expressed in lung adenocarcinoma tissues ,and lipoprotein lipase ( LPL) gene is obvious down-regulated in lung adenocarcinoma tissuesResult:1. The gene expression were obvious difference between lung cancer tissues and the para-neoplastic tissues,36 genes differential expression of lung squamous cell carcinoma (LSCC) were screened out,comprising 23 known genes, 13 unknown genes, among these genes, up and down regulated genes were 22 and Irrespectively.And,259 genes associated with the lung adenocarcinoma were found,containing 137 known genes, 122 unknown genes, among them,146genes were up-regulated and 113genes were down-regulaed. The small-cell lung cancer had 95 genes differential expression, containing 56 known genes, 39 unknown genes, among these genes, up and down regulated genes were 70 and 25.2. We discovered for the first time that Metallothionein-3 gene ( MT-3) is highly expressed in lung adenocarcinoma tissues,which was up-regulated 5 times than in the normal lung tissues.And lipoprotein lipase ( LPL) gene is obvious down-regulated in lung adenocarcinoma tissues, The data of cDNA microarray hybridization and Northern blots showed that LPL gene was down-regulated 4 times in the tissues of adenocarcinoma.conclusion:1. The gene expression were obvious difference between lung cancer tissues and the para-neoplastic tissues, Those genes changes involved...
